摘要 目的研究没食子酸乙酯(EG)对高转移性乳腺癌细胞株黏附、侵袭和迁移能力影响及其作用机制。方法观察EG干预后高转移性乳腺癌MDA-MB-231与对照组相比黏附、侵袭、迁移能力的变化,采用SABC法和实时PCR法检测EG对丝/苏氨酸激酶(AKT-2)和e IF-4E的蛋白表达及mRNA表达的影响。结果加入EG干预的高转移性乳腺癌细胞株的黏附、侵袭和迁移能力均低于对照组(P﹤0.05);EG组的AKT-2和e IF-4E蛋白表达及mRNA表达量均低于对照组,两组间差异有统计学意义(P﹤0.05)。结论体外EG能降低MDA-MB-231的侵袭能力,可能与抑制癌细胞中原癌基因和延长因子蛋白以及mRNA的表达有关。 Objective To study the effect of ethyl gallate (EG) on the adhesion, invasion and migration ability of in high-ly metastatic breast cancer cell line. Method The changes of adhesion, invasion and migration ability of MDA-MB-231 af-ter EG treatment were observed, while SABC and real-time PCR were applied to detect the effect of EG on protein and mRNA expression of serine/threonine kinase (ATK-2) and eIF-4E. Result The adhesion, invasion and migration ability of highly metastatic breast cancer cell line with EG treatment was weaker than that without EG (P〈0.05), besides, the pro-tein and mRNA expression of AKT-2 and eIF-4E in EG group was lower than that in control group, with significant differ-ence observed (P〈0.05). Conclusion In vitro EG treatment reduces the invasion ability of the MDA-MB-231 cancer gene, which may be related to the inhibited expression of proto-oncogene, elongation factor and mRNA.
