桑瓜饮降血糖作用的有效部位筛选 被引量:5

Active Parts Screening of Hypoglycemic Effects of Sanggua Drink

摘要 目的观察桑瓜饮对血糖、血脂的调节作用,筛选出其具有降血糖作用的有效部位。方法高脂饮食联合链脲佐菌素构建2型糖尿病大鼠模型,造模成功的糖尿病大鼠随机分为9组,即B组(模型组)、C组(阳性药物组)、D组(桑瓜饮水煎剂组)、E组(水液部位组)、F组(乙酸乙酯部位组)、G组(正丁醇部位组)、H组(乙酸乙酯部位+正丁醇部位组)、I组(乙酸乙酯部位+水液部位组)、J组(正丁醇部位+水液部位组),正常组记为A组,灌胃给药,每日1次,给药42d。末次给药后,检测并比较桑瓜饮水煎剂及其各部位对2型糖尿病大鼠血糖、血脂指标的调节作用。结果造模后给药前,与A组相比,各组大鼠的空腹血糖(FBG)显著升高(P<0.01),差异具有统计学意义;B组和各给药组之间无显著性差异(P>0.05)。给药42d后,与B组相比,C、D、E、J组的FBG显著降低(P<0.05或P<0.01),FBG下降百分比分别为96.75%、17.48%、24.10%、66.90%;与C组相比,除J组外,其余给药组FBG均与C组在显著性差异(P<0.01)。血脂检测指标方面,与B组相比,C、G、H、J组均有降低甘油三酯(TG)的作用(P<0.05或P<0.01),TG含量分别下降88.10%、49.06%、51.92%、88.10%;C、D、H、J组有降低总胆固醇(TC)的作用(P<0.05或P<0.01),其含量分别下降17.90%、12.22%、12.64%、17.44%;升高高密度脂蛋白胆固醇(HDL-C)含量的分别是D组和J组(P<0.05),HDL-C含量分别升高30.15%和37.10%;桑瓜饮各给药部位均有降低低密度脂蛋白胆固醇(LDLC)水平的作用(P<0.05或P<0.01);与C组比较,J组TC、TG、LDL-C均与之无显著性差异,但其HDL-C含量比C组升高13.33%。结论桑瓜饮对2型糖尿病大鼠的血糖血脂均有调节作用,其有效部位可能主要是正丁醇部位+水液部位。 Objective To screen the active parts of hypoglycemic effects of Sanggua drink. Methods Rats models of type 2 diabetes induced by high fat diet combined with streptozotocin were randomly divided into 9 groups,namely B( model group),C( positive drug group),D( Sanggua drink decoction group),E( aqueous parts group),F( ethyl acetate fraction group),G( n- butanol fraction group),H( a mixture of ethyl acetate fraction and n- butanol fraction group),I( a mixture of ethyl acetate fractionand aqueous parts group),J( a mixture of n- butanol fraction and aqueous parts group). A is the serial number of the normal group. The drugs were given by intragastric administration for 42 d,once per day. At the late of medication,compare the regulation of each part of Sanggua drink on the indexes of blood glucose and blood lipids of type 2 diabetic rats. Result Before administration,compared with A,the levels of fasting blood glucose( FBG)were significantly different; FBG between B and each treatment group showed no significant difference. After administration,compared with B,the level of FBG of C,D,E and J significantly decreased,the decline in percentage were 96. 75%,17. 48%,24. 10%,66. 90%. Compared with C,in addition to J,FBG of other treatment groups were significantly different. Compared with B,the levels of triglyceride( TG)of C,G,H,and J significantly decreased,the decline in percentage were 88. 10%,49. 06%,51. 92%,88. 10%. The levels of total cholesterol( TC) of C,D,H,J decreased,the decline in percentage were 17. 90%,12. 22%,12. 64%,17. 44%. The content of high- density lipoprotein cholesterol( HDL- C) increased in D and J,the percentage were 30. 15% and 37. 10%,and the content of low- density lipoprotein cholesterol( LDL- C) decereased in each group of Sanggua drink. Compared with C,there were no significant difference in content of TG,TC,LDL- C of J,but the content of HDL- C of J increased by 13. 33%. Conclusion Sanggua drink has a regulatory effect on blood glucose and blood lipid in type 2 diabetic rats,in which a mixture of n- butanol fraction and aqueous parts of the group is the most effective part.
