补肾活血中药体外诱导子宫内膜干细胞凋亡和抑制Wnt信号通路激活的实验研究 被引量:1

摘要 目的:研究补肾活血中药体外诱导对子宫内膜干细胞以及Wnt信号通路激活的影响。方法:选取子宫内膜异位症患者和健康人为样本,通过分离培养其在位、异位子宫内膜的干细胞;分别予以补肾活血中药、西药后对各组子宫内膜干细胞凋亡指数以及Wnt通路因子的表达。结果:高剂量补肾活血以及西药均可诱导子宫内膜干细胞凋亡,并可降低子宫内膜干细胞Wnt7a、β-catenin、VEGF、MMP-9通路因子的表达,分离的蛋白表达有意义。结论:子宫内膜干细胞凋亡功能下降以及Wnt信号通路激活是子宫内膜异位症的发病机制,补肾活血中药可促进子宫内膜干细胞发生凋亡,从而起到治疗的作用。 Objective: To study the effect of Chinese herbal medicine on inducing endometrial stem cells apoptosis and inhibiting the activation of Wnt signaling pathway in vitro. Methods: Health and endometriosis patients were selected to isolate and culture the ectopic endometrial stem cells. After intervention by Chinese and Western medicine respectively,test each endometrial stem cells’ apoptosis index and the Wnt pathways factor expression. Results: After drug intervention,it found that high dose of kidney- invigorating and blood circulation- promoting Chinese drugs and Western medicine can induce apoptosis of endometrial stem cells and can reduce the expressions of Wnt7 a,β- catenin,VEGF and MMP- 9 in endometrial stem cells. Conclusion: The apoptosis of endometrial stem cells and the activation of Wnt signaling pathway are the pathogenesis of endometriosis and Chinese herbs can promote the apoptosis of endometrial stem cells,which can play a role in the treatment.
