脊髓NMDA受体NR2B参与前列腺炎致跨器官痛觉致敏的机制研究

摘要 目的研究N-甲基-D-天冬氨酸(NMDA)受体NR2B在慢性前列腺炎性刺激所致的跨器官痛觉过敏中的作用。方法采用完全氟氏佐剂前列腺注射制作慢性前列腺炎大鼠模型,用逆转录-聚合酶链反应和蛋白质印迹技术分别检测NMDA受体NR2B亚基m RNA及蛋白表达水平的变化,以及鞘内给予NR2B特异性拮抗剂——ifenprodil对NR2B表达的影响。结果慢性炎性刺激后腰骶段脊髓背角NMDA受体NR2B表达上调,鞘内给予ifenprodil后NR2B在脊髓背角表达下调。结论 NMDA受体NR2B可能参与了慢性前列腺炎导致的跨器官痛觉致敏,特异性NR2B拮抗可能是临床治疗慢性前列腺炎导致的慢性神经病理性痛的潜在策略。 Objective To investigate the role of N-methyl-D-aspartate(NMDA) receptor NR2 B in the cross-organ pain sensation sensitization caused by inflammation stimulation of chronic prostatitis. Methods The prostatic injection of complete Freund adjunvant(CFA) was adopted to establish the rat chronic prostatitis model. The changes of NR2 B subunit m RNA and pro-tein expression levels were detected by using the Western blot and RT-PCR methods. Furthermore,the influence of NR2 B specific antagonist ifenprodil by intrathecal injection on NR2 B expression was investigated. Results The expressions of NR2 B in the spinal dorsal horn of lumbosacral segment after chronic inflammatory stimulation was up-regulated,while which after the intrathecal administration of ifenprodil was down-regulated. Conclusion The NMDA receptor NR2 B may be involved in the cross-organ pain sensation sensitization caused by chronic prostatitis. Therefore specific NR2 B may be a potential strategy for treating chronic neuropathic pain induced by chronic prostatitis.
