不同处理对红心杉微扦插苗木生根及叶绿素荧光参数的影响 被引量:7

Effects of Various Stroma and Root Agent on Seedling Rooting and Chlorophyll Fluorescence Parameters in Cunninghamia lanceolata

摘要 以广西红心杉产地的优良红心杉单株组培苗为试验材料,采用L9(34)正交试验设计,研究不同栽培基质、生长调节剂种类及浓度对红心杉组培苗微扦插生根率、平均生根数量、平均根长、叶绿素含量及叶绿素荧光参数等指标的影响,探讨叶绿素、叶绿素荧光参数与生根的关系,为优化和筛选红心杉组培苗微扦插培养条件提供参考.结果表明:影响红心杉微扦插苗木生根率、叶绿素含量的主要因素依次为IBA浓度、基质类型、NAA浓度.IBA对平均根长、生根率、叶绿素总含量及F_o,ΦPSⅡ的影响较大,不同水平的IBA对叶绿素总含量和ΦPSⅡ的影响有显著差异;NAA对F_m,F_v/F_o,F_v/F_m及Yield的影响较大,不同水平的NAA对Yield的影响有显著差异;基质对保存率及平均根数量的影响较大,不同类型基质对各指标的影响均不大.F_o,ΦPSⅡ与生根率呈显著正相关,ΦPSⅡ与叶绿素总含量呈极显著正相关.综合评定结果显示:处理5(IBA 50 mg/L+NAA 25 mg/L处理苗木,黄心土为基质)为红心杉微扦插育苗的最佳组合,各项指标表现均较均衡. Effects of various stroma and root agent on seedling rooting and chlorophyll fluorescence parameters in Cunninghamia lanceolata were studied by using L9(34) orthogonal experimental design ,with the excellent clones of Cunninghamia lanceolata tissue culture seedlings. The rooting rate, average root number, average root length, content of chlorophyll and chlorophyll fluorescence parameters were measured. The relationship of chlorophyll, chlorophyll fluorescence parameters and rooting were investigated in order to provide reference for the optimization and selection of Cunninghamia lanceolata micro cutting. The results showed:The main factors affecting seedling rate and content of chlorophyll of Cunninghamia lanceolata micro cutting were concentration of IBA, substrate types and concentration of NAA. IBA had a great influence on the average root length, rooting rate, total chlorophyll content, Fo and φPS Ⅱ. Different levels of IBA had significant difference on the total chlorophyll content and φPS Ⅱ. NAA had a great influence on Fm, Fv/Fo, Fv/Fm and Yield. Different levels of NAA had significant difference on Yield. Matrix had a great influence on preservation rate and average root number, and different types of matrix had little effect on the index. Fo, φPS Ⅱand rooting rate were significantly positively correlated.φPS Ⅱ and chlorophyll content showed highly significant positive correlation. Comprehensive evaluation results showed that treatment 5, used IBA 50 mg/L and NAA 25 mg/L to treat seedlings, and used yellow subsoil as the matrix, was the most conducive to Cunninghamia lanceolata micro cutting. In the treatment, the indicators are more balanced.
