半夏厚朴汤加味对慢性萎缩性胃炎大鼠胃黏膜血流量及脂质过氧化损伤的影响研究 被引量:22

摘要 目的观察半夏厚朴汤加味对慢性萎缩性胃炎(CAG)模型大鼠胃黏膜血流量及脂质过氧化损伤的影响,探讨其作用机制。方法随机选取18只大鼠作为正常组,90只大鼠用N-甲基-N’-硝基-N-亚硝基胍制作CAG模型。将造模成功后CAG大鼠随机分为模型组、维酶素组、半夏厚朴汤加味高剂量组、半夏厚朴汤加味中剂量组、半夏厚朴汤加味低剂量组,每组18只。维酶素组给予维酶素0.28 g/kg灌胃,半夏厚朴汤加味高、中、低剂量组分别给予半夏厚朴汤水煎剂10.0 g/kg、5.0 g/kg、2.5 g/kg灌胃,均2 m L/只,1次/d,连续10周。检测各组大鼠胃黏膜血流量、胃液游离酸度和总酸度及胃蛋白酶活性,胃黏膜组织中超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)活性。结果模型组大鼠胃黏膜血流量、胃液游离酸度和总酸度、胃蛋白酶活性及胃黏膜组织中SOD、GSH-Px活性均显著低于正常组(P均<0.05),胃黏膜组织中MDA活性明显高于正常组(P<0.05);维酶素组和半夏厚朴汤各剂量组大鼠胃黏膜血流量、胃液游离酸度和总酸度、胃蛋白酶活性及胃黏膜组织中SOD、GSH-Px活性均显著均显著高于模型组(P均<0.05),MDA活性均明显低于模型组(P均<0.05),且高、中剂量组效果优于低剂量组(P均<0.05)。结论半夏厚朴汤加味可能通过增加胃黏膜血流量、调节胃酸和胃蛋白酶分泌及抗脂质过氧化损伤而起保护和修复胃黏膜作用。 Objective It is to observe the effect of Banxia Houpo Decoction (BHD) on blood flow and lipid peroxidation injury in gastric mucosa in rats with chronic atrophic gastritis(CAG), and to investigate the mechanism. Methods 18 rats were randomly selected as normal group, another 90 rats were selected to establish CAG model with N methyl N nitro N nitrosog- uanidine. The successful models were randomly divided into model group, Vitacoenzyme group treated with Vitacoenzyme 0.28 g/kg, high, medium and low dose of BHD groups treated with 10.0, 5.0, 2.5 g/kg BHD respectively, 18 rats in each group. All the rats were treated once per day and for 10 weeks. After 10 weeks, secretion of gastric acid and pepsin activity, gastric mu- cosal blood flow(GMBF) , the SOD, MDA and GSH in gastric mlucosa were observed or detected. Results The secretion of gastric acid and pepsin activity and the levels of GMBF, SOD, GSH - Px in gastric mucosal were significantly lower while that of MDA was higher in model group than those in normal group ( P all 〈 0.05 ). The secretion of gastric acid and pepsin activity and the levels of GMBF, SOD, GSH - Px in gastric mucosal were significantly higher while that of MDA was lower in Vitaco- enzyme group and BHD groups than those in model group (P all 〈0. 05) , and the decrease or increase were more significant in high dose group and medium group than that in low dose group (P 〈 0.05). Conclusion Banxia Houpo Decoction has good curative effect on CAG, its mechanism may be related with improving blood circulation of gastric mucosa, strengthening gastric mucosl protective screen, regulating gastric acid and pepsin activity, resisting the free radical imp airment and so on.
