摘要
目的:探讨姜黄素对大鼠心脏缺血再灌注损伤的作用及机制。方法:通过夹闭大鼠左冠状动脉45min,再灌注2h构建心脏缺血再灌注损伤模型。检测各组大鼠肌酸激酶(CK)、乳酸脱氢酶(LDH)、活性氧自由基(ROS)、线粒体膜电位、三磷酸腺甙(ATP)、BCL-2、Bax、BCL-XL、Caspase 3、Cleaved-Caspase 3、Caspase 9、Cleaved-Caspase 9和细胞质中AIF和cytochrome C的表达水平、心脏病理形态和血流动力学。结果:姜黄素可减轻心脏病理和血流动力学改变,减少CK、LDH、ROS、Bax、Cleaved-Caspase 3、Cleaved-Caspase 9和细胞质中AIF和cytochrome C的表达,但可增加线粒体膜电位、BCL-2、BCL-XL、Caspase 3和Caspase 9和ATP的表达。结论:姜黄素预处理可通过抑制ROS介导的线粒体凋亡信号通路途径减轻心脏缺血再灌注损伤。
Objective:To explore the effect and mechanism of Curcumin on myocardial ischemia reperfusion injury.Method:The model of myocardial ischemia reperfusion injury was induced by clamping the left anterior descending coronary artery for 45 min and reperfusion for 2hour in rats.The rats of Cardiac pathological morphology,hemodynamics,ATP concentration,mitochondrial membrane potential and the expression of CK-MB,LDH,ROS,BCL-2,Bax,BCL-XL,Caspase3,Cleaved-Caspase3,Caspase9,Cleaved-Caspase9 and cytoplasmic AIF and cytochrome c were evaluated in all groups.Result:Curcumin can significantly decrease the change of hemodynamics and Cardiac pathological morphology.Curcumin reduced the expression of CK,LDH,ROS,Bax,CleavedCaspase3,Cleaved-Caspase9,cytoplasmic AIF and cytochrome c and up-regulated the mitochondrial membrane potential,the expression of BCL-2,BCL-XL,Caspase3,Caspase9 and ATP concentration.Conclusion:Curcumin pretreatemnt can protect rats against myocardila ischemia-reperfusion injury by suppressing ROS-mediating mitochondrial apoptosis.
