摘要
目的研究脯氨酸羟化酶2(prolyl hydroxylase2,PHD2)在鼻咽癌组织和癌旁组织中的表达差异及其临床意义。方法运用免疫组化染色及评分、荧光定量PCR方法检测56例鼻咽癌组织及56例癌旁组织中PHD2的表达差异,分析PHD2表达水平与鼻咽癌患者临床病理指标之间的关系。结果免疫组化结果显示,在56例鼻咽癌组织中PHD2阳性率19.6%(11/56),其中"-"为45例,"+"为6例,"++"3例,"+++"2例;56例癌旁组织中PHD2阳性率为69.6%(39/56),"-"为17例,"+"为8例,"++"10例,"+++"21例,鼻咽癌组织的PHD2表达水平显著低于癌组织(P<0.05);荧光定量PCR检测结果显示,在鼻咽癌组织中PHD2 mRNA的表达水平低于癌旁组织,差异具有统计学意义(P<0.05)。PHD2的表达与患者年龄、性别、分化程度无相关性(P>0.05),而与鼻咽癌的临床分期、T分期、淋巴结转移具有相关性(P<0.05)。结论 PHD2在鼻咽癌的发生发展中可能起到抑制癌症进展的作用,患者的PHD2表达水平可能与其分期及转移有关。
Objective To explore the expression of prolyl hydroxylase 2 ( PHD2 ) and its clinical significance in nasopharyngeal carcinoma and adjacent tissues. Methods Immunohistochemistry and Real time PCR were used to detect PHD2 expression level of 56 cases of nasopharyngeal carcinoma and adjacent tissues, and to analysis the relationship between PHD2 expression and clinic pathological features of nasopharyngeal carcinoma. Results In 56 cases of nasopharyngeal carcinoma, the positive rate in carcinoma was 19.6% (11/56)," -" 45 cases," + " 6 cases," ++ "3 cases," ++++ " 2 cases; and the positive rate in adjacent tissues was 69.6% (39/56) o Real time PCR showed PHD2 expression level of nasopharyngeal carcinoma was significantly lower than that in adjacent tissues(P 〈 0.05 ). CCR1 expression had no correlation with age, sex and degree of differentiation ( P 〉 0.05, but it correlated with clinical stage, T stage, development of nasopharyngeal carcinoma(P 〈 0.05 ). Condusion PHD2 is negatively expressed in the majority of NPC, and its expression level may be related to the patient's prognosis.
