摘要
目的观察消痰通腑方对肥胖小鼠体重及脂代谢的影响,探讨其作用机制。方法将34只小鼠随机分成正常对照组(n=12)和肥胖造模组(n=22)。用高脂饲料建肥胖模型,11周后,两组各处死6只小鼠,检测体重、体脂、三酰甘油(TG)、总胆固醇(TC)、白细胞介素-6(IL-6)及脂联素(ADP)水平。将剩余的肥胖造模组小鼠(n=16)剔除体重最轻的4只小鼠后,随机分为肥胖模型组(n=6)及消痰通腑方组(n=6),消痰通腑方小鼠组予消痰通腑方灌胃,正常对照组及肥胖模型组均灌胃等体积蒸馏水,每日1次,时间为4周。实验结束后分别检测小鼠体重、体脂、TG、TC、IL-6及ADP水平。结果肥胖造模组小鼠第11周体重、体脂及TC、IL-6水平均显著高于正常对照组,血清ADP水平显著低于正常对照组(P<0.01)。消痰通腑方组小鼠体重、体脂显著低于肥胖模型组(P<0.05),TC及IL-6水平低于肥胖模型组(P<0.05),血清ADP水平高于肥胖模型组(P<0.05)。结论消痰通腑方能降低肥胖模型小鼠的体重、体脂、TC及IL-6水平,其机制可能与增加血清ADP水平有关。
Objective To observe the effect of Xiaotan Tongfu Prescription on weight and lipid metabolism in obese mice, and to explore its mechanism. Methods 34 mice were randomly divided into the normal control group (n=12) and the obese model group (n=22). High-fat diet was given to establish obese modeling. After 11 weeks, 6 mice of each group were sacrificed, to test body weight, body fat, triglyceride (TG), total cholesterol (TC), interleukin-6(IL-6) and adiponectin (ADP). The remaining obese model group mice (n=16) to eliminate the lightest weight of 4 mice were randomly divided into the obese model group (n=6) and Xiaotan Tongfu Prescription treatment group (n=6). Drugs were given to the mice in Xiaotan Tongfu Prescription treatment group once a day for 4 weeks, the mice in other groups were given the same volume of distilled water. In the ending, the mice were killed and body weight, body fat, TG, TC, IL-6 and ADP were all detected and analyzed. Results IN 11 weeks, the body weight, body fat and TC, IL-6 of the obese model group were significantly higher than those of the normal control group, serum ADP level was significantly lower than that of the normal control group (P 〈 0.01). The body weight, body fat of the Xiaotan Tongfu Prescription treatment group were significantly lower than those of the obese model group (P 〈 0.05), TC and IL-6 levels were lower than those of the obese model group (P 〈 0.05), serum ADP level was significantly higher than that of the obese model group (P 〈 0.05). Conclusion Xiaotan Tongfu Prescription can reduce the body weight, body fat, TC and IL-6 levels of the obese model mice, the mechanism may be associated with the increased ADP level.
