Kruppel-like factor 5 (KLF5) is an evolutionarily conserved zinc finger transcrpfion factor that regulates the expressions of many downstream genes. It takes part in the regulations of proliferation, differentiation and apoptosis. Post-translational modifications influence the stability of KLF5 and its roles. In addition, there are interactions between KLF5 and non-coding RNAs (ncRNAs). Increasing researches showed that the expression of KLF5 was frequently abnormal in human cancers including colorectal cancer, breast cancer, prostate cancer and so on. KLF5 involves in carcinogenesis and is associated with prognosis of patients with cancer. The function of KLF5 reveals context-dependent in some types of cancer. Post-translational modifications and sex hormone always affect its function. Due to its significant roles in human cancers, KLF5 may be a potential diagnostic biomarker and therapeutic target for human cancers.
