摘要 目的探讨FSP1在小鼠巨噬细胞中的表达特征及其在巨噬细胞功能的意义。方法以FSP1^-GFP小鼠作为研究对象,采用流式细胞染色检测FSP1在小鼠巨噬细胞中的表达情况;应用多种细胞因子对FSP1^-巨噬细胞进行刺激,观察FSP1的表达情况;应用过氧化氢(H_2O_2)对巨噬细胞进行氧化应激诱导,并观察其FSP1的表达情况及上清中FSP1的浓度;流式检测FSP1^-和FSP1^+的巨噬细胞细胞因子分泌情况及吞噬功能。结果腹腔巨噬细胞和骨髓诱导来源的巨噬细胞均可被分为FSP1^+和FSP1^-两群。随着H_2O_2刺激腹腔巨噬细胞时间延长,FSP1^+巨噬细胞的比例下降,ELISA检测上清中的FSP1浓度增加;刺激FSP1^+和FSP1^-两群巨噬细胞对LPS诱导的IL-6、IL-12、TNF-α等促炎性细胞因子无明显差异;FSP1^-的腹腔巨噬细胞对鸡红细胞及乳胶微球的吞噬能力高于FSP1^+巨噬细胞。结论依据巨噬细胞对FSP1的表达可将巨噬细胞分为FSP1^+和FSP1^-两群。FSP1^+巨噬细胞经H_2O_2刺激分泌大量FSP1蛋白。这两群巨噬细胞对促炎性细胞因子的分泌无明显差异,但FSP1^-巨噬细胞的吞噬能力高于FSP1^+巨噬细胞。 Objective To determine the expression of fibroblast-specific protein 1(FSP1) on macrophages and its roles on macrophage functions.Methods FSP1-GFP reporter mice was used in the present study. Macrophages from FSP1^-GFP mice were treated with H_2O_2 and the FSP1 expression was detected by flow cytometry and ELISA in protein level. The expressions of cytokines and the phagocytosis on FSP1^- or FSP1~+ macrophages were assayed by flow cytometry. Results Peritoneal macrophages and bone marrow-derived macrophages can be divided into either FSP1~+ or FSP1^- subpopulations. The percentage of FSP1~+ macrophages was decreased and FSP1 in the supernatant was increased after oxidative stress induction by H_2O_2 stimulation. There was no significant difference between FSP1^- and FSP1~+ macrophages on the expressions of pro-inflammatory cytokines like IL-6, IL-12 and TNF-α after stimulated with LPS. The phagocytosis on chicken red blood cells and latex beads of FSP1^-macrophages was higher than FSP1~+ macrophages. Conclusions Macrophages can be divided into two subpopulations depending on the expression of FSP1. FSP1~+ macrophages can secret abundant FSP1 protein after H_2O_2 stimulation. There is no significant difference between the two subpopulations of macrophages on the secret of cytokines, but FSP1^- macrophages own higher phagocytosis ability than FSP1~+ macrophages.
