不同诱导培养基和籼粳成分对水稻花药培养效果的影响 被引量:2

Effects of Induction Medium and Indica-japonica Composition on Rice Anther Culture

摘要 以5份不同籼粳成分的水稻为试验材料,分别以不同基本培养基(改良M8,SK3,合5,NB)、碳源(蔗糖和麦芽糖)、激素(单一激素、复合激素)为变量,设计16种诱导培养基进行花药培养,探讨水稻适宜的花培条件。结果表明,不同材料花药培养力差异显著,零轮、蜀恢527花药培养力显著高于9311/广粳1号∥9311、9311/广粳1号∥蜀恢527、9311/零轮;籼型基因频率与绿苗率呈显著负相关(-0.89*);基本培养基GM_8和合5花培效果较好,NB培养基不适合供试材料的花药培养;以蔗糖为碳源,添加复合激素的处理花培效果较好。改良的M_8+50 g/L蔗糖+2 mg/L 2,4-D+1 mg/L NAA+1 mg/L KT是适宜水稻花药培养的诱导培养基。 Using five rice materials with different indica-japonica composition as the test materials,the optimal anther culture conditions were studied by designing 16 induction media based on basic culture media( modified M8,SK3,He 5,NB),carbon sources( sucrose and maltose) and plant hormones( single and composite). The results showed that there existed significant difference in anther culture ability among the tested rice materials,in which Linglun and Shuhui 527 had a significantly higher anther culture ability than the other three materials,i. e.,the BC1F1 of 9311 / Guangjing 1 / /9311,the F1 of 9311 / Guangjing 1 / / Shuhui 527 and the F1 of 9311 / Linglun. The frequency of indica component( Fi) of rice materials was negatively correlated to the green plantlet rate with the coefficient at-0. 89 significant at the 5% level. Among the basic culture media,GM8 and He 5 had better anther culture effects,but NB was not suitable for the tested rice materials. The induction media with addition of sucrose as the carbon source plus composite hormones had better anther culture effects. In this study,the optimal induction medium for anther culture of rice was the modified M8+ 2 mg / L 2,4-D + 1 mg / L NAA + 1 mg / L KT.
