丝胶对2型糖尿病大鼠胰岛素水平的影响

摘要 目的:观察丝胶对2型糖尿病模型大鼠血清胰岛素水平的影响。方法:48只雄性SD大鼠随机分为正常对照组、2型糖尿病模型组、丝胶治疗组和二甲双胍阳性对照组4组,每组12只大鼠。采用高脂高糖饲料喂养联合链脲佐菌素(35mg/kg,2次)连续腹腔注射的方法制作2型糖尿病大鼠模型,以空腹血糖≥11.1mmol/L作为成模标准。待模型成功建立后,丝胶组和阳性对照组大鼠分别给予丝胶(2.4g/kg/d)和二甲双胍(55.33mg/kg/d)灌胃35天。分别采用葡萄糖氧化酶法和ELISA法检测各组大鼠的血糖和血清胰岛素水平。结果:模型组大鼠的血糖明显高于正常组大鼠、血清胰岛素水平明显低于正常组大鼠(P<0.05);丝胶治疗组、阳性对照组大鼠的血糖明显低于模型组大鼠,血清胰岛素水平明显高于模型组大鼠(P<0.05)。结论:丝胶可提高2型糖尿病大鼠血清胰岛素水平,这可能是丝胶降血糖作用的机制之一。 Objective: To observe the effects of sericin on serum insulin level of type 2 diabetic rats. Methods: 48 male SD rats were randomly divided into 4 groups with 12 rats in each group: normal control group, type 2 diabetes mellitus model group, sericin treatment group and metformin positive control group. The type 2 diabetes mellitus rats model was induced by high calorie and high sugar diet plus intraperitoneal injection of streptozotocin(35mg/kg), and the standard for successfully establishing type 2 diabetes model was fasting blood glucose≥11.1mmol/L. Then the rats in sericin treatment group and postitive control group were respectively lavaged with sericin(2.4g/kg/d) and metformin(55.33mg/kg/d) for 35 days. Glucose oxidase method and ELISA were used to measure blood glucose and serum insulin level respectively. Results: The blood glucose of rats in model group increased obviously compared with rats in normal group, serum insulin level decreased obviously(P 0.05); The blood glucose of rats in sericin treatment group and positive control group declined obviously compared with rats in model group, serum insulin level rised obviously(P 0.05). Conclusions: Sericin can increase the serum insulin level of diabetic rats, this maybe one of the mechanisms that sericin decreasing blood glucose.
