摘要
目的:利用HPLC法对丹参配方颗粒中丹参酮ⅡA的实际含量进行精确测定.实验条件:选用高效液相色谱法测定,分离条件为Agilent Zorbax SB-C18色谱柱(250mm×4.6mm,5μm),甲醇-水(75:25)为流动相,流速1.0 m L/min,检测波长为270nm.结果:丹参酮ⅡA的线性范围为8.03~41.0μg·m L-1(r=0.999 2),平均加样回收率为97.3%(RSD=1.04%).结论:HPLC法不仅便于操作、且灵敏度强,在调控丹参配方颗粒含量方面发挥相关作用.
Objective: In order to establish an accurate and practical HPLC method for determinating the math of Tanshinone IIA in Salvia Miltiorrhiza Granules the HPLC methocl is used. Methods: Chromatographic separation was operated on an Agilent Zorbax SB- Cl8 column( 250 mm × 4. 6mm,5μm) with mobile phase of methanol( It's ratio IS 75: 25),water solution at a flow rate of 1. 0 m L / min and a column temperature of 22℃. The eluted analytes were detected by on- 1ine UV detector whose wavelength was set at 270 nm. Results: The calibration curves of Tanshinone IIA was linear over the ranges of 8. 03 ~ 41. 0 μg /m L- 1( r = 0. 999 2),and the average recoveries was 97. 3%( RSD = 1. 41%). Conclusion: The established method is simple,rapid,reliable and can be used for drug contro1. It is meaningful for using in Clinical practice.
