摘要
目的探讨不同的血液贮存温度和贮存时间对去白细胞悬浮红细胞白细胞清除效果的影响,选择并建立合适的去除白细胞的操作方法,进一步提高血液中白细胞的清除效果。方法抽取本站采集的无偿献血者全血220{9,在不同的贮存温度(2℃~6℃、18℃~25℃)、贮存时间(2、4、6、12、24、36、48、60、72h)和即采即滤条件下随机分组,制备去白细胞悬浮红细胞,分别留取相应的标本检测血液过滤前后的白细胞含量,对检测结果进行对比、分析。结果①2℃~6℃组白细胞残留量符合国家标准且明显低于18℃~25℃组和即采即滤组,差异有统计学意义(P〈0.0i)。②2h组白细胞残留量明显高于4h组,检测结果均符合国家标准,两者比较差异有统计学意义(P〈0.01);4~72h组白细胞残留量呈逐步增高趋势,4~24h各组白细胞残留量均明显低于国家标准,与4h组相比差异无统计学意义(P〉O.05);36h组、48h组白细胞残留量符合国家标准,与4h组相比差异有统计学意义(36h组:P〈0.05;48h组:P〈0.01);60h组、72h组白细胞残留量超过国家标准,与4h组相比差异有统计学意义(P〈0.01)。结论全血采集完成应置于4℃4-2℃环境条件下贮存4h后,最好在24h内但不超过48h,尽快制备去白细胞悬浮红细胞,可有效提高白细胞清除效果。
Objective To investigate the influences caused by blood storage temperatures and time which affect the leukocytes reduced of red blood cells in additive solution leukocytes. We also aimed at selecting and establishing proper operation methods of leukocytes reducing which will further improve the effects of leukocytes reduced in blood. Methods Extracting 220 portions of voluntary blood donors' whole blood collected by our department. Making randomized grouping under different storage temperatures(2℃-6℃, 18℃-25℃), different storage times(2 h, 4 h, 6 h, 12 h, 24 h, 36 h, 48 h, 60 h, 72 h) and filtering immediately to produce red blood cells in additive solution leukocytes reduce and taking leukocytes contents of the specimens detected blood which were filtered and unfiltered to contrast and analysis the detection results. Results (1)The leukocytes residues in group 2℃~6℃ are compliant with the national standards and apparently lower than leukocytes residues in group 18℃~25℃ and 37℃, the differences was statically significant(P〈0.01). (2)The leukocytes residues in group 2h are apparently higher than leukocytes residues in group 4h and the detection results are all compliant with the national standards , the differences was statically significant (P〈0.05).The leukocytes residues in 4h group to 72 h group show a gradually increasing trend. The leukocytes residues in group 4 h to 24b are all apparently lower than the national standard, and comparing with group 4h, the differences was not statically significant (P〉0.05). The leukocytes residues in group 36h ,group 48h are compliant with the national standard, and comparing with group 4h, the differences was statically significant (Group 36 h: P〈0.05, group 48 h: P〈0.01). The leukocytes residues in group 60h, group 72 h are higher than the national standards, and comparing with group 4h, the differences was statically significant (P〈0.01). Conclusion Whole blood collection should be put into an 4℃±2℃ environment to store for 4 hours, and it is better to produce red blood cells in additive solution leukocytes reduced earlier, within 24 hours but no more than 48 hours, which can effectively improve the effects of leukocytes reduce.
