大黄鱼C型溶菌酶对鳗弧菌胁迫的响应研

摘要 溶菌酶作为是鱼体内非特异性免疫因子,在机体起到消化和防御病毒作用。本研究克隆了大黄鱼C型溶菌酶基因ORF长度为333bp,可编码110个氨基酸,Gen Bank数据库的登陆号为XM-010753143.1,相对分子质量为27.68k Da,理论等电点为5.24,具有8个半胱氨酸,能形成4对二硫键,有2个保守的催化残基位点Glu17和Asp34。多序列比对显示,大黄鱼C型溶菌酶氨基酸序列与其他鱼类同源性高达71%以上。进化树发现,大黄鱼C型溶菌酶首先与鲈形目鱼类聚在一起,鱼类C型溶菌酶与哺乳动物的亲缘关系近。基因结构分析可知,大黄鱼C型溶菌酶基因含有3个外显子和2个内含子。实时荧光定量PCR检测显示,大黄鱼C型溶菌酶8种组织(肝脏、脾脏、肾脏、脑、心、肌肉、鳃、肠)中均有表达,其中在肝中表达水平量最高(800多倍),脾和肾组织最低。鳗弧菌和poly I:C感染后,大黄鱼肝脏、脾脏和肾脏组织中C型溶菌酶基因的表达变化不大,说明C型溶菌酶在大黄鱼体内主要起到消化作用。
