益气健脾方对甲状腺功能减退大鼠海马CA1区神经颗粒素及LTP的影响

Effect of Chinese Medical Prescription of Strengthening Spleen and Replenishing Qi on Neurogranin and LTP of Hypothyroidism Rats Hippocampus CA1 Region

摘要 目的:观察益气健脾方对甲状腺功能减退大鼠海马CA1区神经颗粒素(Neurogranin,Ng)及长时程增强(long-term potentiation,LTP)损伤的影响。方法将SPF级成年Wistar大鼠,每日灌服他巴唑混悬液2.7 mg·kg-1,持续45 d。第46天,将造模后大鼠随机分为6组:正常对照组、模型组、左旋甲状腺素片组、益气健脾方小剂量组、益气健脾方大剂量组、益气健脾方合甲状腺片组,除正常对照组和模型组灌胃10 m L·kg-1去离子水,其余各组灌胃不同药物,1次·d-1,共15d。观察大鼠海马CA1区Ng及LTP的表达。结果各治疗组可显著改善甲状腺功能减退症大鼠海马CA1区Ng蛋白表达水平,与模型组比较,差异均有显著性(P<0.01),左甲状腺素片组、益气健脾大剂量组、益气健脾方加左甲状腺素组等3组之间无显著性差异(P>0.05),左甲状腺素片组、益气健脾大剂量组、益气健脾方加左甲状腺素组与益气健脾小剂量组比较,优于益气健脾小剂量组,差异有统计学意义(P<0.01)。甲状腺功能减退大鼠,经过左旋甲状腺素及益气健脾方的治疗后的LTP幅度明显高于模型组(P<0.01),但是甲状腺素片组、益气健脾大剂量组、益气健脾方加左甲状腺素组与益气健脾小剂量组的LTP幅度仍低于正常对照组的LTP幅度(P<0.05)。结论益气健脾方在一定程度上可修复甲状腺功能低下引起的海马CA1区的LTP的损伤,能显著提高大鼠脑C1区Ng水平。 Objective: To observe the effects of Chinese medical prescription of strengthening spleen and replenishing Qi on neurogranin and LTP of hypothyroidism rats hippocampus CA1 region. Method: To give SPF level adult rats antithyroid drugs( ATD)and produce hypothyroidism models. Drug administration: Tapazole suspension 2. 7 mg·kg- 1per day for 45 days. On day 46,randomly group rats into 6 groups,the control group,the model group,the levothyroxine sodium tablets group,the small dosage strengthening spleen and replenishing Qi prescription group,the large dosage strengthening spleen and replenishing Qi prescription group,strengthening spleen and replenishing Qi prescription and thyroxine combination group. Given the control group and model group 10 mL ·kg- 1deionized water every day,and given other groups different drugs,once per day for 15 days,and observed the effects on neurogranin and LTP of hypothyroidism rats hippocampus CA1 region. Results: The protein expression level of hypothyroidism rats CA1 region of every group was dramatically improved,and there was a significant difference between that of the model group( P〈0. 01),meanwhile,there was no significant difference among the levothyroxine sodium tablets group,the large dosagestrengthening spleen and replenishing Qi prescription group,and strengthening spleen and replenishing Qi prescription and the thyroxine combination group( P〈0. 05); the effects of the levothyroxine sodium tablets group,the large dosage strengthening spleen and replenishing Qi prescription group,and strengthening spleen and replenishing Qi prescription and the thyroxine combination group was better than that of the small dosage strengthening spleen and replenishing Qi prescription group,there was a significant difference( P〈0. 01). After the treatment of levothyroxine sodium tablets and strengthening spleen and replenishing Qi prescription,the LTP of pothyroidism rats was observably higher than that of the model group( P〈0. 01),but LTP of levothyroxine sodium tablets group,the small dosage strengthening spleen and replenishing Qi prescription group,the large dosage strengthening spleen and replenishing Qi prescription group,strengthening spleen and replenishing Qi prescription and the thyroxine combination group was still lower that that of control group( P〈0. 05). Conclusion: Strengthening spleen and replenishing Qi prescription can mend the LTP impairment on hippocampus CA1 region caused by hypothyroidism,and can significantly improve the Ng level of hypothyroidism rat brains.
