抑癌基因PTEN甲基化和胃癌的生物学关系探究 被引量:1

摘要 目的对比胃癌组织与癌旁正常组织中抑癌基因PTEN的甲基化情况,分析基因甲基化与胃癌之间的生物学关系。方法选取2013年8月~2015年7月我院收治的43例胃癌患者,采用免疫组化方法对入选患者的胃癌组织与癌旁组织的PTEN甲基化情况进行检测并对比,分析甲基化和胃癌临床分期、转移的关系。结果胃癌组织的PTEN蛋白阳性率低于癌旁正常组织(P〈0.05),组间对比差异具有统计学意义(χ2=8.49,P〈0.05);且胃癌组织的PTEN甲基化阳性率高于癌旁组织,且高、中分化腺癌的甲基化阳性率低于低分化腺癌,差异均具有统计学意义(P〈0.05)。结论抑癌基因PTEN甲基化与胃癌的淋巴结转移存在相关性,且PTEN的蛋白表达水平与胃癌的临床病理分期有密切关系,可将PTEN视作胃癌生物学行为的主要评价指标。 Objective To compare the gastric carcinoma and adjacent normal tissue methylation of tumor suppressor gene PTEN and analysis of biological relationships between gene methylation in gastric cancer. Methods From August 2013 to July 2015,43 cases of patients with gastric cancer were selected,the application of immunohistochemical methods detected on the gastric carcinoma and paracancerous tissue of patients and comparing the methylation of PTEN status,analysis of the association of methylation and clinical stages of gastric cancer and metastasis. Results The positive rate of PTEN in gastric carcinoma normal tissue was lower than that of normal adjacent mucosa(P〈0.05),and the difference was statistically significant between the groups(χ2= 8.49,P〈0.05),and methylation of PTEN-positive gastric cancer rates were significantly higher than that of normal adjacent mucosa,and the methylation rate of high and medium differentiation adenocarcinoma were significantly lower than those of poorly differentiated adenocarcinoma,the difference was statistically significant(P〈0.05). Conclusion The methylation of tumor suppressor gene PTEN and correlation of lymph node metastasis in gastric cancer,and PTEN protein expression levels have close relation with clinical pathological staging of gastric cancer,PTEN can be seen as the main evaluation indexes of biological behavior in gastric carcinoma.
