姜黄素抑制膀胱癌T24细胞增殖及Hsp90α的表达

Effects of Curcumin on Inhibiting the Proliferation of Bladder Cancer T24 Cells and Hsp90α Expression

目的探讨姜黄素对膀胱癌T24细胞增殖以及热休克蛋白90α(Hsp90α)表达的影响。方法培养T24细胞株,分别用不同浓度姜黄素(0,5,10,15,20μmol/L)处理细胞24小时和20μmol/L姜黄素分别处理细胞不同时间(0,6,12,24,48小时),采用四唑盐(MTT)比色法,检测T24细胞的生长抑制率,采用逆转录聚合酶链式反应(RT-PCR),检测Hsp90α基因mRNA的表达,以蛋白免疫印迹法(Western blotting),检测Hsp90α蛋白质的表达。结果姜黄素处理后的T24细胞代谢MTT能力降低,细胞生长抑制率明显增加;细胞Hsp90α基因mRNA和蛋白质的表达均下调,并且这些效应呈浓度和时间依赖性(P<0.05)。结论姜黄素可能通过调控Hsp90α的表达而抑制T24细胞的增殖。

Objective To investigate the effect of curcumin in cell proliferation and expression of heat shock protein 90 alpha （Hsp90a） in bladder cancer cells. Methods Culture of bladder cancer cell line （T24 cell line） ,The cells were treated with different concentrations of curcumin （0,5,10,15,20 μmol/L） for 24 hours,or 20 mol/L curcumin were treated with 0,6,12,24,48 h,respectively.Then the growth inhibition rate of T24 cells was detected by the colorimetric assay of four thiazole salt （MTT） ,the expression of Hsp90 mRNA was detected by reverse transcription polymerase chain reaction （RT-PCR） ,and the expression of Hspg0 protein was detected by Western blotting. Results Compared with the control group, the metabolism ability of T24 cells decreased and the cell growth inhibition rate was increased significantly after the curcumin treatment.At the same time, the expression of Hsp90 mRNA and protein in T24 cells was down regulated, and these effects were concentration and time dependent （P〈0.05）. Conclusion Curcumin may inhibit the proliferation of bladder cancer cells by regulating the expression of Hsp90.
