摘要
建立了一种同时测定四季三黄丸中盐酸小檗碱、黄芩苷和栀子苷含量的反相高效液相色谱(RP-HPLC)法.色谱条件为Agilent Zorbax SB-C18分离柱(250 mm×4.6 mm,粒径5μm),乙腈-6 mmol/L KH_2PO_4流动相梯度洗脱,流速1.0m L/min,多波长切换检测.结果表明,盐酸小檗碱、黄芩苷和栀子苷线性范围分别为3.8 28.4μg/m L(r=0.999 8),16.5 123.8μg/m L(r=0.999 6)和3.8 28.4μg/m L(r=0.999 7),加样回收率分别为97.2%(RSD=1.5%),97.7%(RSD=2.6%)和102.9%(RSD=2.8%).本方法可以同时分析四季三黄丸中盐酸小檗碱、黄芩苷和栀子苷含量,对该制剂质量进行有效监控.
A new RP-HPCL method was developed for the simultaneous determination of berberine hydrochloride,baicalin and geniposide in Siji Sanhuang Pills. The chromatographic separation was carried out on a Agilent Zorbax SB-C18 column(250 mm ×4.6 mm,5 μm)using 6 mmoL/L KH2PO4-acetonitrile as mobile phase at a flow rate of 1. 0 mL/min under gradient elution. The quantitative linear range was 3. 8~28. 4 μg/mL for berberine hydrochloride,16. 5~123. 8 μg/mL (r=0. 999 6) for baicalin and 3. 8~28. 4 μg/mL(r=0. 999 7 ) for geniposide, respectively. The average recoveries were 97. 2% ( RSD =1. 5%) for berberine hydrochloride,97. 7% (RSD=2. 6%) for baicalin and 102. 9% (RSD=2. 8%) for geniposide,respectively. This method can simultaneously analysis the contents of berberine hydrochloride,baicalin and geniposide in Siji Sanhuang Pills,and can effectively control the quality of Siji Sanhuang Pills.
