原儿茶酸对Huh7细胞中HNF1α表达的抑制影响 被引量:2

摘要 研究原儿茶酸(0,0.1,1,10μg/m L)处理Huh7细胞24 h和48 h对肝细胞核因子1α(HNF1α)表达的影响.荧光定量PCR和蛋白印迹检测结果表明,原儿茶酸不仅能抑制Huh7细胞中HNF1α的mRNA表达水平,也能抑制Huh7细胞中HNF1α的蛋白表达水平.由于HNF1α可以调控乙肝病毒的S1和C启动子活性,本研究结果提示,原儿茶酸可能通过抑制肝细胞HNF1α的表达而发挥抗HBV作用. Hepatocyte nuclear factor 1α( HNF1α) is an important regulator of glucose and amino acid homeostasis in hepatocytes and plays an important role in liver development, differentiation and normal liver structure maintenance. In this study,we examined the effect of protocatechuic acid ( PA) (0,0. 1,1,10 μg/mL) for 24 h or 48 h on HNF1α expression in Huh7 cells. The results indicate that PA can not only inhibit the mRNA of HNF1α but also suppress the protein of HNF1α in Huh7 cells evidenced by real?time PCR and western blot. Since HNF1α can regulate the activity of hepatitis B virus preS1 and core promoter,our results indicate that PA can inhibit HBV replication in vitro by inhibiting HNF1α expression.
