下调雌激素受体相关受体α对不同类型子宫内膜癌细胞增殖凋亡的影响 被引量:1

Influence of proliferation and apoptosis caused by estrogen receptor-related receptorαdown-regulation in different types of endometrial cancers

摘要 目的探讨雌激素受体相关受体α(Estrogen receptor-related receptorα,ERRα)特异性拮抗剂XCT790作用于不同类型子宫内膜癌细胞时ERRα的表达改变及细胞增殖、凋亡情况。方法将XCT790药物作用组设为XCT组,未加药物处理组设为阴性对照组(Negtive Control,NC),加入XCT790溶剂DMSO设为空白对照组(Control,CON),比较ERRαmRNA表达改变。采用定量PCR(Quantitative PCR,qPCR)和免疫印迹法(Western blot,WB)分别检测RL952、AN3-CA[(Estrogen receptorα+,ERα+),ERRα+]和HEC-1A、HEC-1B(ER-,ERRα+)细胞的ERRαmRNA和蛋白表达水平;采用XCT790处理4株子宫内膜癌细胞,qPCR检测ERRαmRNA的表达情况,WB检测ERRα蛋白的表达情况。应用噻唑蓝比色法(MTT)及流式细胞术分别检测4株子宫内膜癌细胞生长和凋亡情况。结果RL952、AN3-CA细胞ERRαmRNA表达量高于HEC-1A、HEC-1B细胞,差异有统计学意义(P<0.05)。WB显示,ERRα蛋白在RL952、AN3-CA中均呈较高表达,而HEC-1A、HEC-1B中均呈较低表达,与mRNA水平相吻合。XCT790处理后,XCT组与CON组相比ERRαmRNA和蛋白表达水平均显著下降,差异有统计学意义(P<0.05)。NC组与CON组ERRαmRNA和蛋白表达差异无统计学意义(P>0.05)。MTT法显示,肿瘤细胞数量在XCT790作用下显著减少,差异有统计学意义(P<0.05);流式细胞术显示各株细胞的XCT790组凋亡率较各自CON组显著升高,差异有统计学意义(P<0.05)。结论 XCT790能有效抑制子宫内膜癌中ERRα的表达水平,ERRα表达下调与子宫内膜癌细胞增殖、凋亡密切相关。 Objective To explore XCT790, the specific antagonist of estrogen receptor-related receptor s, affecting the expression of ERRs and cell proliferation as well as apoptosis in different types of endometrial cancer cells . Methods Cells were treated with 10-5M XCT790 (DMSO soluble, Sigma) as XCT group , DMSO as a control group and no drug as negtive group. Four Endometrial cancer cells mRNA and protein expression level of ERRs were detected by qPCR and western blot respectively. XCT790 affected the cells, and the expression level of ERRs in four endometrial cancer cells above were detected by qPCR and western blot respectively. Then, the changes of cell proliferation and apoptosis were measured by MTT and FCM. Results The ERRs mRNA expression levels of ERRs in RL952 and AN3-CA were significantly higher than those in HEC-A and HEC-1B (P 〈0.05). A similar result was observed in the protein expression of ERRα. After XCT790 affecting the cells, endometrial cancer cells rnRNA and protein expression in the NC group had no differences with CON group in each cell (P〉0.05). But the XCT group had remarkable lower ERR mRNA and protein expression than CON group in each cell (P〈0.05). MTT showed the numbers of cell were significantly reduced by XCT790 in each cell (P〈0.05). FCM described the apoptosis ratio of CON group was outstanding lower than XCT group in each cell (P〈0.05). Conclusion XCT790 can inhibits the expression of ERRα in different types of endometrial cancer cells. ERRα low expressing has a close relationship with cell proliferation and apoptosis in endometrial cancer cells.
