激光多普勒血流仪在改良大鼠颈动脉狭窄模型中的应用 被引量:1

摘要 目的建立一种改良的大鼠颈动脉狭窄模型,并用激光多普勒血流仪监测术中及术后颈动脉血流变化情况。方法 18只雄性SD大鼠随机分为假手术组、模型组,假手术组大鼠仅切开皮肤,模型组大鼠行颈部胸锁乳突肌前缘斜切口做颈总动脉球囊损伤术以成模,并用激光多普勒血流仪在术中及术后14天观察颈动脉血流情况,并取损伤侧颈总动脉,行HE染色,观察术后第1天及14天后血管内皮损伤及内膜增生情况。结果模型组大鼠颈动脉球囊损伤术后颈动脉血流明显减少,血管内皮损伤严重;术后14天颈动脉血流有所增加,但仍较假手术组少(P<0.05),血管内膜增生明显、管壁变厚。结论改良手术方法能够更好地建立大鼠颈动脉狭窄模型,激光多普勒血流仪能够有效地观察到颈动脉血流变化情况。 Objective To establish a modified model of rat carotid artery stenosis, and monitor artery blood flow changes of intraoperative and postoperative and with laser Doppler flowmetry. Methods Eighteen male SD rats were randomly divided into sham operation group and model group: the rats in sham operation group only underwent skin incision, and the rats in model group underwent carotid artery balloon injury surgery by cervical thoracic lock mastoid muscle front oblique. Laser Doppler flowmetry was used in intraoperative and postoperative 14 days to observe the blood flow. Then all the rats were killed, and H. E staining was used to observe vascular endothelial injury and intimal hyperplasia. Results The blood flow of model group rat after carotid artery balloon injury was significantly reduced, with severely vascular endothelial injury; the carotid artery blood flow of postoperative 14 days increased, but still less compared to the sham operation group (P〈0.05), and the endometrial hyperplasia and thickness were obvious in model rats. Conclusion A rat model of carotid artery stenosis can be well established by the modified surgical method, and laser Doppler flowmetry can effectively observe blood flow changes of carotid artery.
