摘要
随着造血干细胞(HSC)移植技术的发展,脐血已成为干细胞的主要来源之一。为突破HSC来源及数量不足的制约,HSC体外扩增技术发展迅速,涉及到细胞因子、小分子化合物的应用以及联合培养及三维培养技术的发展和氧环境在干细胞扩增技术的作用以及基因修饰技术。本文就近年上述技术中采用的主要方法进行综述。
With the development of hematopoietic stem cell transplantation technology, umbilical cord blood has become one of the main sources of hematopoietic stem cells (HSC). Significant efforts have gone into developing technologies for ex vivo expansion of HSC in order to overcome the limited number of HSC. A variety of approaches for HSC ex vivo expansion are reviewed in the article, such as cytokine, small molecule compounds, aerobic environment, gene modification technique, co-culture and three-dimensional cell culture technology.
