酶解法制备岩藻黄素及其对不同来源糖苷酶抑制作用的研究 被引量:2

摘要 目的:本研究主要是为了探讨岩藻黄素的提取纯化方法及其对不同来源糖苷酶的抑制作用。方法:本研究以大连产海带为原材料,通过酶解辅助提取法及硅胶柱层析柱法制备岩藻黄素,并测定其对酵母来源的α-葡萄糖苷酶与大鼠肠来源的蔗糖酶和麦芽糖酶抑制作用。结果:通过单因素实验与正交实验确定了岩藻黄素最佳的酶解制备条件:纤维素酶和果胶酶添加量分别为7.5%和3%(酶容积/海带干重质量),酶解pH为5.0,酶解温度为50.0℃,酶解时间1.5 h,提取溶剂为80%乙醇,液料比为1∶15,提取温度为55℃,提取时间为24 h,岩藻黄素得率为1.471±0.063 mg/g(以干重计算)。经过大量提取、收集、硅胶柱层析纯化后的岩藻黄素纯度达到了92.33%,且其对酵母来源的α-葡萄糖苷酶与大鼠肠来源的蔗糖酶、麦芽糖酶均具有较弱的抑制活性,说明岩藻黄素的虽具有体外降糖作用,但是效果不显著。 Purpose:The purpose of this study is to investigate the extraction method and the hypoglycemic effect( in vitro)of fucoxantin. Method :In this study,the fucoxanthin was extracted from seaweed using enzymatic extraction and silica gel chromatography purification method. Results :The optimal extraction conditions were :7.5% cellulose concentration( w/w),3% pectinase concentration( w/w),enzymolysis temperature 50℃,enzymolysis time1.5h,enzymolysis pH 5.0,extraction solvent 80% ethanol,solid to liquid ratio 1 ∶ 15,extraction temperature 55℃,extraction time 24 h. Under the above conditions,the fucoxanthin content was 1.471 ± 0.063 mg/g( dry weight). After silica gel chromatograph,92.33% purified fucoxanthin was obtained and test showed that it had weak inhibitory activities on a various glycosidase.
