基于prrnS基因序列分析黑斑蛙裂头蚴种系发育关系

摘要 为研究湖南省黑斑蛙裂头蚴分离株的核糖体(rDNA)小亚基基因(rrnS)部分序列(prrnS)的遗传变异情况,并用所获得prrnS序列构建黑斑蛙裂头蚴与其他绦虫的种群遗传关系。利用聚合酶链反应(PCR)扩增湖南省黑斑蛙裂头蚴的prrnS,将PCR产物进行测序并对其序列进行分析。9株湖南省黑斑蛙裂头蚴分离株的prrnS序列长度均一致,为330 bp。经分析显示,黑斑蛙裂头蚴prrnS序列种内相对保守,种间差异较大,故可作为蛙裂头蚴的种间遗传变异研究的分子遗传标记。 The purposes of the present experiment are to examine sequence variation in the small subunit ribosomal RNA (rrnS) gene of F.sparganum isolates from Hunan province, and to construct their phylogenetie relationship based on rrnS sequences. The partial rrnS (prrnS) was amplified from each F. sparganum sample by PCR and PCR product was sequenced. The results showed that the lengths of prrnS sequence were 330 bp. The results indicated that there was no significant variation in rrnS sequences within F.sparganum, while intra-species difference was obvious. It was concluded that prrnS sequences could be used as genetic marker for population genetic studies of eestodes.
