摘要 对1株耐盐枯草芽孢杆菌TGBio-1433培养基、培养条件进行了优化;为了获得更高的菌体浓度,还探索了枯草芽孢杆菌TGBio-1433补料分批发酵工艺。试验结果表明,最优培养基为:玉米粉10 g/L、豆粕粉20 g/L、葡萄糖15g/L、蛋白胨15 g/L、牛肉膏5 g/L、MgSO41 g/L、CaCO37 g/L。最优培养条件为:起始pH 7.0-7.5,发酵温度35℃,培养时间18-20 h,接种量5%补料分批发酵工艺为:发酵16 h后,一次性补加碳、氮源(葡萄糖∶蛋白胨∶牛肉膏=3∶3∶1)总量的15%。枯草芽孢杆菌TGBio-1433发酵液活菌数由最初的24.2×10^8cfu/m L提高到136.1×10^8cfu/mL。 Fermentation medium and conditions of Bacillus subtilis TGBio- 1433 were investigated,in order to obtain a higher concentration of bacteria,the fed-batch fermentation of Bacillus subtilis TGBio- 1433 were also explored. The results showed that the optimal combination for fermentation was as follows: corn flour 10 g / L,soybean meal 20 g / L,glucose 15 g / L,peptone 15 g /L,beef paste 5 g / L,MgSO4 1 g / L,CaCO3 7 g / L. The optimum for fermentation conditions was as follows: the initial pH 7.0 -7.5,incubation temperature 35 ℃,incubation time18 - 20 h,inoculums concentration 5%. Fed-batch fermentation process was as follows: after 16 h fermentation,adding carbon and nitrogen source( glucose ∶ peptone ∶ beef paste = 3 ∶ 3 ∶ 1) 15% of the total one-off. The concentration of Bacillus subtilis TGBio- 1433 from the original 24.2× 10^8 cfu / mL up to 136.1× 10^8 cfu / mL.
