医用丝素蛋白皮肤再生膜的生物相容性评价 被引量:6

摘要 背景:皮肤再生膜是与皮肤缺损表面接触的生物材料,必须考虑其对机体的毒性。目的:综合评价丝素蛋白皮肤再生膜的生物相容性。方法:(1)皮肤刺激实验:在新西兰大白兔皮肤缺损处分别敷贴医用丝素蛋白皮肤再生膜、涂抹甲醛、涂抹生理盐水,24,72 h后观察红斑和水肿情况。(2)急性全身毒性实验:通过小鼠尾静脉分别注射医用丝素蛋白皮肤再生膜材料浸提液、苯酚及生理盐水,注射后24,48,72 h,观察小鼠状态、毒性反应和死亡。(3)细胞毒性实验:分别以100%、50%、25%、10%浓度的材料医用丝素蛋白皮肤再生膜材料浸提液培养L-929细胞,培养后2,4,7 d,用紫外分光光度仪检测吸光度值;培养48 h,MTT法检测吸光度值。(4)以100%浓度的材料医用丝素蛋白皮肤再生膜材料浸提液培养L-929细胞,48 h后采用q PCR法检测细胞纤维连接蛋白m RNA表达。结果与结论:丝素蛋白皮肤再生膜对皮肤缺损创面无不良反应,未对机体皮肤产生刺激,对机体无急性毒性作用,无细胞毒性,不影响体内细胞纤维连接蛋白的表达,具有较好的生物相容性。 BACKGROUND: Skin reproducing membrane is a biomaterial used directly in contact with skin defects, so its toxicity to the body must be taken into consideration. OBJECTIVE: To systematical y evaluate the biocompatibility of skin reproducing membrane of medical fibroin. METHODS: Skin irritation test: the skin defect of New Zealand white rabbit was covered with the skin reproducing membrane of medical fibroin, formaldehyde or normal saline, respectively, and erythema and edema were observed at 24 and 72 hours after treatment. Acute systematic toxicity test: the mice were given extracts of skin reproducing membrane of medical fibroin, phenol and normal saline via tail vein injection; then status of mice, toxicity grade and death were recorded at 24, 48 and 72 hours after injection. Cytotoxicity test: L-929 cel s were co-cultured with 100%, 50%, 25%, 10% skin reproducing membrane of medical fibroin extracts, and absorbance values were detected by ultraviolet spectrophotometry at 2, 4 and 7 days after culture, as wel as by MTT assay after 48 hours of culture, respectively. Besides, L-929 cel s were co-cultured with 100% skin reproducing membrane of medical fibroin extracts, and mRNA expression of fibronectin was determined by qPCR technology after 48-hour culture. RESULTS AND CONCLUSION: Skin reproducing membrane of medical fibroin has no adverse reaction, acute cytotoxicity, skin irritation and systemic toxicity, and additional y, it does not affect mRNA expression of fibronectin. In general, skin reproducing membrane of medical fibroin has good biocompatibility.
