摘要
目的探讨XLOC_004122在结核患者支气管肺泡灌洗液的表达以及功能,评价长链非编码RNA(XLOC_004122)作为诊断MTB感染新标志物的潜力。方法收集解放军第309医院2014年10月-2015年3月结核科48例患者支气管肺泡灌洗液标本,结合患者的临床症状、体征、实验室检查、影像学检查等将其分为结核病组和非结核对照组,应用实时荧光定量PCR检测XLOC_004122的差异表达;通过基因GO功能注释、富集分析和KEGG信号转导通路富集分析,阐明XLOC_004122靶基因参与调控的细胞功能与信号通路。结果与非结核对照组相比,结核病组XLOC_004122的表达量显著下调(t=-2.487,P<0.01);GO分析结果显示XLOC_004122靶基因功能主要富集于离子通道的活性、通道复合物的合成、跨膜转运活性等过程;KEGG信号转导通路主要富集于Ca2+、toll样受体等信号通路,通过XLOC_004122共表达的Ca2+通道信号通路可以看出其在NCX、OPCR、CaV1、PTK、MLCK和CAMK中均有差异表达。结论 XLOC_004122在结核感染中呈相对低表达,且可能与结核病的发生有关,可尝试将其作为诊断结核感染的新指标。
Objectives To investigate the differential expression and function of long non-coding RNA(XLOC_004122)in bronchoalveolar lavage fluid from patients with tuberculosis and to preliminarily evaluate whether XLOC_004122could be a new marker for diagnosis of a Mycobacterium tuberculosis infection. Methods A total of 48 samples of bronchoalveolar lavage fluid were collected from PLA Hospital 309 from October 2014-March 2015.In accordance with data such as clinical manifestations,signs,laboratory tests,and imaging studies,samples were divided into those from patients infected with M.tuberculosis and a control group.Real-time quantitative PCR was used to assess XLOC_004122in macrophages from patients infected with M.tuberculosis and the control group.Cell functions and signaling pathways regulated by the target gene(XLOC_004122)were determined using GO and KEGG pathway analysis. Results XLOC_004122was found to be significantly down-regulated in patients infected with M.tuberculosis compared to the control group(t=-2.487,P〈0.01).GO analysis indicated that the function of XLOC_004122is mainly to enhance ion channel activity,formation of channel complexes,and transmembrane transport.KEGG pathway analysis indicated XLOC_004122enriches the Ca2+signaling pathway,the Toll-like receptor pathway,and other signal pathways.In addition,co-expression network analysis indicated that XLOC_004122 was differentially expressed in NCX,OPCR,CaV1,PTK,MLCK,and CAMK of the Ca2+channel signaling pathway. Conclusion XLOC_004122is expressed at low levels in patients with tuberculosis,and this expression may be associated with the pathogenesis of tuberculosis.XLOC_004122may be a novel index with which to diagnose tuberculosis.
