摘要
目的 观察葡萄籽原花青素(GSPE)和长波紫外线(UVA)对人角质形成细胞(HaCaT细胞)增殖的影响,以探讨GSPE有无光保护作用。方法 MTT法分别测定不同浓度GSPE以及不同剂量UVA对HaCaT细胞增殖活性的影响。结果 GSPE浓度为5-200μg/m L时,对HaCaT细胞增殖影响不明显,浓度为500μg/m L及以上时抑制细胞生长,且有统计学意义;UVA剂量超过10 J/cm2有抑制作用且随剂量增高抑制作用越明显,且有统计学意义,25 J/cm2时其抑制率达37.65%;浓度为50、100μg/mL的GSPE对25 J/cm2 UVA照射后的HaCaT细胞的增殖有明显促进作用。结论25 J/cm2的UVA对HaCaT细胞的增殖有明显抑制作用,而合适浓度的GSPE对HaCaT细胞增殖活性有光保护作用。
Objective In order to observe the effects of GSPE and UVA on the proliferation of HaCaT cells, and to investigate the light protection of GSPE. Methods The cells proliferation were detected by MTr method. Results The impact on HaCaT cells proliferation was not obvious when the concentration of GSPE was lower than 200 μg/mL. But when the concentration of GSPE was more than 500 μg/mL could significantly inhibit the growth of the cells. It could inhibit the growth of HaCaT cells when the dose of UVA was more than 10 J/cm^2. And the 25 J/cm^2 dose of UVA could inhibit the proliferation of HaCaT cells to 37.65%. The concentration of 50 μg/mL and 100 μg/mL of GSPE could promote the proliferation of HaCaT cells which were irradiated by 25 J/cm^2 dose of UVA. Conclusion The 25 J/cm^2 dose of UVA could inhibit the proliferation of HaCaT cells significantly. And appropriate concentration of GSPE can play a role in light protection.
