摘要
采用膜技术对蛋白质进行分离纯化已受到生命科学领域的广泛关注,而智能开关膜为蛋白质精细分离提供了新的解决方案.本文以接枝甲基丙烯酸-N,N-二甲氨基乙酯(DMAEMA)的乙烯-乙烯醇共聚物(EVAL)分离膜为智能开关膜,对其开关特性与蛋白分离性能进行了研究.结果表明,EVAL接枝膜渗透通量具有显著的开关性,且pH交替重复6个单元以后开关性能保持稳定.在模拟蛋白分子牛血清白蛋白(BSA)与溶菌酶(LZM)及混合模拟蛋白分子BSA与LZM分离过程中,接枝率为15%的EVAL接枝膜在pH 3.6的条件下对BSA与LZM分离效果最佳,分离系数分别为9.7和7.8.
Membrane technique used for protein separation has gained great attention in the life science field.Intelligent switch membranes provide an efficient method for efficient protein separation.In this research,intelligent switch ethylene vinyl alcohol copolymer(EVAL)membranes were prepared by grafting dimethylaminoethyl methacrylate(DMAEMA)using UV irradiation graft polymerization.The pH responsive and protein separation properties of the intelligent switch membranes were investigated.The results indicated that the pH-responsive"on/off"properties of the grafted EVAL membrane are observed.During six cyclic measurements,the pH-responsive "on/off"properties are also observed.During the separation of bovine serum albumin(BSA)/lysozyme(LZM)model solution and BSA/LZM binary mixture,the highest separation factoris achieved at pH 3.6of poly(DMAEMA)-grafted EVAL membrane with GD15%.The separation factor values are 9.7and 7.8,respectively.
