丹参破壁饮片、传统饮片对大鼠心肌缺血和血液流变学的影响及急性毒性研究

Comparative Study of Effects between Ultrafine Granular Powder and Traditional Decoction Piece of Salvia miltiorrhiza Bge.on Experimental Myocardial Ischemia and Hemorheology and Their Acute Toxicity

目的:研究、比较丹参破壁饮片、传统饮片改善大鼠心肌缺血和血液流变学的药效作用及急性毒性差异。方法:采用注射垂体后叶素复制大鼠急性心肌缺血模型,比较丹参破壁饮片、传统饮片对大鼠急性心肌缺血的作用;复制大鼠血瘀模型,采用凝血酶诱导血小板聚集,比较丹参破壁饮片、传统饮片对血瘀模型大鼠血液流变学的影响;采用最大给药量法比较丹参破壁饮片、传统饮片对小鼠的急性毒性作用。结果:丹参破壁饮片、传统饮片在注射垂体后叶素3min后均能明显对抗心肌缺血所致心电图T波抬升(P<0.01),二者均能减少大鼠心肌缺血范围(P<0.01),其中丹参破壁饮片低剂量的药效作用与传统饮片相似;丹参破壁饮片、传统饮片均能降低血瘀大鼠的全血黏度和血浆黏度(P<0.01或P<0.05),有效降低凝血酶所致的血小板聚集率(P<0.01)。丹参破壁饮片、传统饮片以一日最大给药量120g/kg灌胃,实验小鼠均无明显急性毒性反应。结论:丹参破壁饮片、传统饮片均对大鼠急性心肌缺血有明显保护作用及改善血瘀大鼠血液流变学的药效,丹参破壁饮片在低于传统饮片的剂量即可发挥与传统饮片相似的药效;丹参破壁饮片和传统饮片用药安全性相似。

Objective：To compa re the difference of effects between ultrafine granular powder and traditional decoction piece of Salvia miltiorrhiza Bge.on improving myocardial ischemia and hemorheology and to compare their acute toxicity.Methods：Acute myocardial ischemia rat model was established by injected with pituitrin,on which effect between ultrafine granular powder and traditional decoction piece of S.miltiorrhiza on acute myocardial ischemia was compared.Blood-stasis rat model was established and platelet aggregation was induced with thrombin,on which effect between ultrafine granular powder and traditional decoction piece of S.miltiorrhiza on hemorheology was compared.Acute toxicity of ultrafine granular powder and traditional decoction piece of S.miltiorrhiza on mice was compared with method of maximum dosage test.Results：Both ultrafine granular powder and traditional decoction piece of S.miltiorrhiza inhibited the elevation of T wave in electrocardiogram of acute myocardial ischemia rats 3 min after injected with pituitrin（P〈0.01）.Both reduced the range of ischemic myocardia（P〈0.01）.Low dosage of ultrafine granular powder of S.miltiorrhiza showed similar effect on myocardial ischemia with traditional decoction piece.Both ultrafine granular powder and traditional decoction piece of S.miltiorrhiza reduced blood viscosity and plasma viscosity in blood stasis rats（P〈0.05,P〈0.01）.Both reduced the platelet aggregation rate induced by thrombin（P 0.01）.Neither mice administered with maximum dosage at 120 g/kg of ultrafine granular powder nor traditional decoction piece of S.miltiorrhiza showed acute toxicity response.Conclusion：Both ultrafine granular powder and traditional decoction piece of S.miltiorrhiza have protective effect on ischemic myocardia rats and improving effect on hemorheology in blood-stasis rats.Ultrafine granular powder of S.miltiorrhiza at lower dosage shows similar effect with traditional decoction piece.The safety of ultrafine granular powder of S.miltiorrhiza is similar with traditional decoction piece.
