摘要
目的:研究氟离子植入猪骨源羟基磷灰石对破骨样细胞体外生物学行为的影响。方法:化学浸泡结合高温烧结法制备氟化猪骨源羟基磷灰石,以小鼠单核巨噬细胞株RAW264.7诱导形成破骨样细胞,扫描电镜观察破骨样细胞在材料表面黏附、分化与功能,CCK-8法检测材料上前破骨样细胞的增殖活性,酶活性定量检测细胞内抗酒石酸酸性磷酸酶(Tartrate resistant acid phosphatase,TRAP)活性。结果:破骨样细胞可在氟离子植入猪骨源羟基磷灰石上黏附、分化并发挥吸收功能,材料对前破骨样细胞的增殖活性无明显抑制作用,但可降低破骨样细胞TRAP的表达。结论:氟离子植入猪骨源羟基磷灰石可抑制破骨样细胞的骨吸收作用。
Objective:To evaluate the effect of fluoridation of porcine hydroxyapatite on biological responses of osteoclast-like cells. Method:Fluorinated porcine hydroxyapatite(FPHA)was fabricated from porcine bone by chemical immersion and thermal treatment. Osteoclast-like cells was induced from mouse monocyte-macrophage cell line RAW264.7. Osteoclast-like cells attachment,differentiation and function was observed by scanning electron microscope. The viability of osteoclast-like precursors was determined by cell counting kit-8. Tartrate resistant acid phosphatase(TRAP)activity was detected quantitatively by TRAP assay Kit. Result:Osteoclast-like cells could attach,differentiate and participate in materials resorption on PHA and FPHA disks. No significant inhibiting effect was observed in osteoclast-like precursors viability test. However,a decrease of TRAP activity was detected. Conclusion:FPHA could inhibit the bone resorption function of osteoclast-like cells.
