三种不同方法处理的蝉蜕抗变态反应研究

Experimental Study on Anti-allergy Effects and Mechanism of Three Different Treated Cicadae Periostracum

将蝉蜕超微粉组、蝉蜕水提物组、蝉蜕醇提物组分别进行小鼠被动皮肤过敏反应(PCA)实验、接触性过敏性皮炎(ACD)实验和化合物48/80诱导的小鼠全身过敏反应实验,观察三种不同方法处理的蝉蜕对三种过敏反应的抑制作用,并对PCA、ACD实验中的小鼠过敏部位进行组织切片,光镜下观察肥大细胞的聚集情况.同时制备各给药组的含药血清,检测不同方法处理的蝉蜕对体外培养的小鼠P815肥大细胞脱颗粒及组胺释放的影,比较蝉蜕的超微粉、水提物、醇提物三种不同方法处理得到产物抗变态反应的差异性,探讨其初步机制.结果表明,不同方法处理的蝉蜕对小鼠PCA、ACD和化合物48/80诱导的全身过敏反应均有不同程度的抑制作用,且不同方法处理的蝉蜕制品对体外培养的P815肥大细胞的脱颗粒及组胺释放有一定的抑制作用,但蝉蜕超微粉较其他两种方法处理的蝉蜕作用更明显.

Passive cutaneous anaphylaxis（PCA）,allergic contact dermatitis（ACD）and anaphylaxis induced by compound 48/80 respectively with cicadae periostracum（CP）ultrafine powder,CP aqueous extract,and CP alcohol extract were conducted on male mice to study the anti-allergy effects of different processed CP.The specimens of blue spotted skin from mice in PCA and the pieces of mice ears in ACD were made into sections.Through colorimetric analysis,histamine release by P815 cells cultivated with drug-included serums was measured with O-phthaladehyde（OPA）.Cicadae periostracum ultrafine powder,aqueous extract,and alcohol extract do have inhibitory effects on PCA,ACD and anaphylaxis induced by compound 48/80.And all these CP extracts can inhibit the aggregation of mast cells in the mouse PCA model.Each treated CP may have inhibitory effects on the degranulation and histamine release of P815 cells,but the inhibition of CP ultrafine powder is stronger than the other processed CP.
