番茄红素抑制食管癌细胞EC109增殖及其作用机制的研究 被引量:3

INHIBITION OF ESOPHAGEAL CANCER CELLS PROLIFERATION BY LYCOPENE AND ITS MECHANISMS

摘要 目的探讨番茄红素抑制食管癌细胞EC109增殖及可能的作用机制。方法通过体外细胞培养,采用MTT法检测番茄红素对经PPARγ抑制剂GW9662处理前后的食管癌细胞EC109活力的影响,Western blotting检测番茄红素对PPARγ蛋白表达水平的影响。SPSS17.0软件对结果进行统计学处理。结果番茄红素可抑制食管癌细胞EC109的活力,并呈剂量-效应关系和时间-效应关系(P<0.05),与番茄红素处理组比较,GW9662和番茄红素联合处理组食管癌细胞活力高(P<0.05),而PPARγ蛋白表达水平低(P<0.05),与DMSO对照组比较,番茄红素处理组PPARγ蛋白表达水平高(P<0.01)。结论番茄红素可抑制食管癌细胞EC109的增殖,通过上调PPARγ蛋白的表达是实现其抑制作用的途径之一。 Objective To investigate the effects of lycopene on the proliferation of esophageal cancer EC 109 cells and elucidate its possible molecular mechanisms. Methods Esophageal cancer EC109 cells were cultured with lycopene in vitro. Cells viability was assessed by MTT method with and without peroxisome proliferator-activated receptor-γ (PPARγ) inhibitor GW9662. PPARγ protein expression level was measured by Western Blotting. Statistical analysis was conducted using SPSS17.0 software. Results Lycopene inhibited the viability of EC109 cells in a dose- and time-dependent manner(P〈0.05) Compared with lycopene treatment alone group, the cell viability of the group with the combined treatment of GW9662 and lycopene was higher (P〈0.05), while PPARγ expression level was lower (P〈0.05). PPARγ expression level after the treatment of lycopene was higher than that of the DMSO group (P〈0.01). Conclusion One of the possible molecular mechanisms in suppressing the proliferation of EC109 by lycopene is associated with increasing the expression of PPARγ protein.
