摘要 采用免疫亲和层析净化高效液相色谱(HPLC)法与酶联免疫吸附(ELISA)法测定小麦样品中的脱氧雪腐镰刀菌稀醇。结果显示:HPLC法和ELISA法的标准曲线线性相关性均良好,相关系数分别为0.999和0.994;三水平加标回收率分别为88.9%~92.3%和104.4%~119.4%,相对标准偏差(n=6)分别为1.73%~7.17%和2.25%~9.65%;进行F检验法和t检验法分析,说明两种方法检测结果无显著性差异性。此两种方法均具有回收率高、重现性好、特异性强等特点,ELISA法适合于大批量样品的检测和筛选,HPLC法适合于逐个样品的分析。 The deoxynivalenol in wheat was determined by HPLC with immunoaffinity column purification and ELISA. The results showed that two methods both had good linear relationship and the correlation coefficients were 0.999 and 0.994;The recoveries of the three levels ranged from 88.9% to 92.3% and 104.4% to 119.4%,and relative standard deviations(n=6) ranged from 1.73% to 7.17% and 2.25% to 9.65%;there was no significant difference between the two inspecion F and t methods. The methods of HPLC and ELISA both had the high recovery,good reproducibility and specificity,and the method of ELISA was suitable for the detection and selection of large quantities of samples,while the method of HPLC was suitable for the analysis of one by one.
