摘要
目的观察地氟烷对卵白蛋白致敏的豚鼠高反应气道平滑肌张力的影响。方法 56只雄性豚鼠随机分为7组:正常组(N)、致敏组(S)、致敏对照组(SC)和致敏地氟烷组[致敏0.5 MAC(最低肺泡有效浓度)地氟烷组(SD 0.5)、致敏1.0 MAC地氟烷组(SD 1.0)、致敏1.5 MAC地氟烷组(SD 1.5)和致敏2.0 MAC地氟烷组(SD 2.0)],每组8只。N组与S组用于气道模型评价;SC组、SD 0.5、SD 1.0、SD 1.5及SD 2.0组用于气道平滑肌测定。通过应用卵白蛋白制备致敏豚鼠模型。测量肺阻力,根据肺阻力对不同剂量乙酰胆碱的峰反应值,得到肺阻力剂量反应曲线,评价致敏气道模型。记录离体致敏豚鼠气道平滑肌对10^(-8)mol/L、10^(-7)mol/L、10^(-6)mol/L、10^(-5)mol/L的卡巴胆碱刺激的峰反应值,评价地氟烷对高反应致敏气道的作用。结果与N组相比,S组能显著升高豚鼠的肺阻力变化曲线;S组的肺阻力峰反应值比N组高40%(当乙酰胆碱剂量为6μg/kg时),P<0.05。与SC组相比,SD 0.5、SD 1.0、SD 1.5、SD 2.0组气道平滑肌张力峰值明显下降;卡巴胆碱剂量为10-5mol/L时,SD 1.5和SD 2.0组峰值分别比SC组降低了12.1%和14.0%,比SD 0.5组降低了8.9%和11.0%,P均<0.05。结论地氟烷能够降低卵白蛋白致敏的豚鼠的离体气道平滑肌张力。
Objective To investigate the effects of desflurane on the airway smooth muscle tension in a model of airway hyperresponsiveness,ovalbumin-sensitized guinea pigs. Methods Fifty-six,male guinea pigs were randomly divided into 7 groups: normal group( N),sensitized group( S),sensitized control group(SC) and 4 sensitized desflurane groups( SD 0.5,SD 1.0,SD 1.5 and SD 2.0). The lung resisitance was recorded and the dose-response curves for acetylcholine of lung resistance were used to evaluate the sensitized airway model. The airway smooth muscle tension was measured to evaluate the brochodilator effect of desflurane. Results Compared with N group,the dose-response curves for acetylcholine of lung resistance were elevated significantly in the S group,increased 40% at the acetylcholine dose of 6. 0 μg / kg in the S group( P〈 0. 05). Compared with SC group,the muscle tensions in the SD 0.5,SD 1.0,SD 1.5 and SD 2.0 groups decreased with the increasing concentrations of carbacholine. The peak responses of muscle tension to carbacholine(10-5mol / L) in SD 1.5 and SD 2.0 groups were 12.1% and 14. 0% lower than that in group SC,8.9% and 11.0% lower than that in group SD 0.5,P〈0.05. Conclusion The use of desflurane could decrease the airway smooth muscle tension of sensitized guinea pigs,which was suggested to be one menchnism of bronchodilatoreffect of desflurane.
