DNA损伤修复蛋白XPA表达水平对膀胱癌细胞顺铂耐药的影响 被引量:1

摘要 【目的】探讨DNA损伤修复蛋白XPA表达水平对膀胱癌细胞顺铂耐药的影响。【方法]Western blot法检测不同膀胱癌细胞株XPA蛋白表达水平;shRNA干扰技术筛选出稳定并低表达XPA的膀胱癌细胞株,检测细胞克隆形成率及细胞凋亡率,比较xPA低表达细胞株与XPA正常表达细胞株对顺铂的敏感性。【结果】XPA表达最高的膀胱癌细胞T24,顺铂耐药程度最高;顺铂处理shRNA~T24-XPA细胞后,相比于shRNA—T24-NC,克隆形成率显著降低(P〈0.01),但相反促进细胞凋亡。【结论】膀胱癌化疗耐药可能与XPA水平高表达有关,干预并下调膀胱癌T24细胞XPA蛋白的表达,可降低细胞的克隆形成率,但相反增强顺铂的凋亡诱导率。
