磨盘草石油醚部位改善异丙肾上腺素致大鼠心力衰竭的研究 被引量:6

摘要 目的:观察磨盘草石油醚部位对异丙肾上腺素(ISO)致大鼠心力衰竭的改善并分析可能机制。方法:采用腹腔注射ISO(2mg/kg)4周的方法建立大鼠心衰模型,后分组进行灌胃给药治疗,连续4周。采用放射性免疫法测定大鼠血清中心钠肽(ANP)、血管紧张素Ⅱ(AngⅡ)、醛固酮(Ald)水平,并取心脏组织称重记录,计算心脏指数,观察各组大鼠的心肌病理形态学特征。结果:与正常对照组比较,模型对照组心脏指数明显增加(P<0.01),及血清中ANP、AngⅡ、ALD均显著升高(P<0.05,P<0.01),心肌纤维排列紊乱,血管扩张,可见小灶性纤维瘢痕组织;与模型对照组比较,磨盘草石油醚部位各剂量组血清中指标均出现显著降低(P<0.05,P<0.01)。心肌病理形态学特征方面,低、中剂量组相比不明显,高剂量组有所改善。结论:磨盘草石油醚部位可改善ISO所致的大鼠心衰,其机制可能与降低血清中的ANP、AngⅡ、Ald的水平相关。 Objective: To observe the effects of petroleum ether extract of abutilon indicum on improving heart failure of rats induced by isoproterenol, and explore the possible mechanisms. Methods: The rat models of heart failure were established by intraperitoneal injecting with ISO(2mg/kg) for 4 weeks, and then the rats were grouped and received gavage administration for 4 weeks. The levels of atriopeptin(ANP), angiotensin Ⅱ(Ang Ⅱ), and aldosterone(Ald) in serum of rats were tested by using radioimmunoassay. The weight of heart tissues was weighed and recorded, and the cardiac index(CI) was calculated. The pathological feature of heart tissue was observed. Results: Compared with control group, the CI and levels of ANP, Ang Ⅱ and ALD of rats in model group were all increased significantly(P〈0.05, P〈0.01). The pathological features of heart tissue of rats in model group were showed as disordered arrangement of myocardial fibers, hemangiectasis, and mini-focal fibrous scar tissues. Compared with model group, the serum indexes of rats in various dosage groups of petroleum ether extract of abutilon indicum were reduced to different degree(P〈0.05, P〈0.01), and the CI of rats in high dosage group was superior to that of control. Pathomorphism difference in heart tissue of rats between middle and low dosage groups was not significant, and the improvement in heart tissue pathomorphism of rats in high dosage was significant. Conclusion: Petroleum ether extract of abutilon indicum could improve heart failure of rats induced by isoproterenol, and the mechanism might be related to the lower levels of serum ANP, Ang Ⅱ and Ald.
