固本防哮饮对哮喘缓解期小鼠IL-17A、IL-10和CXCL10的影响 被引量:5

Effects of Guben Fangxiao Decoction on expression of IL-17A, IL-10, and CXCL10 in mice with asthma remission

摘要 目的:探讨中药复方固本防哮饮对哮喘缓解期小鼠白细胞介素-17A(IL-17A)、白细胞介素-10(IL-10)及CXC趋化因子配体10(CXCL10)表达的影响。方法:采用卵蛋白(OVA)致敏和呼吸道合胞病毒(RSV)滴鼻诱导激发BALB/c雌性小鼠,建立哮喘缓解期动物模型,随机分为正常组,模型组,固本防哮饮低、中、高剂量组,孟鲁司特钠组及地塞米松组。用ELISA法和Real-time PCR法分别检测肺组织中IL-17A、IL-10及CXCL10的表达水平。结果:模型组小鼠肺组织中IL-17A、CXCL10表达均显著高于正常组(P<0.01),而IL-10表达显著低于正常组(P<0.01);固本防哮饮低、中、高剂量组,孟鲁司特钠组及地塞米松组小鼠肺组织中IL-17A、CXCL10表达均低于模型组(P<0.05),而IL-10表达高于模型组(P<0.05)。结论:固本防哮饮治疗哮喘的作用机制可能是通过上调IL-10水平,降低IL-17A、CXCL10水平,减轻气道炎性反应,从而达到防治哮喘的目的。 Objective: To investigate the effects of Guben Fangxiao Decoction on expression of interleukin-17A(IL-17A), nterleukin-10(IL-10), and CXC chemokine ligand 10(CXCL10) in mice with asthma remission. Methods: The mouse models with asthma remission were established by using ovalbumin(OVA) sensitization and respiratory syncytial virus(RSV) nasal inhalation in BALB/c female mice. The mice were randomly divided into normal group, model group, montelukast group, dexamethasone group, low, middle, and high dose of Guben Fangxiao Decoction groups. ELISA and Real-time PCR were used to detect the expression levels of IL-17 A, IL-10, and CXCL10 in lung tissues. Results: The expression levels of IL-17 A and CXCL10 in lung tissues of mice in model group were significantly higher than that of normal group(P〈0.01), while the expression of IL-10 was significantly lower than that of normal group(P〈0.01). The expression levels of IL-17 A and CXCL10 in lung tissues of mice in low, middle and high dose of Guben Fangxiao Decoction groups, montelukast group and dexamethasone group were lower than that of model group(P〈0.05), while the expression IL-10 was higher than that of model group(P〈0.05). Conclusion: The mechanism of Guben Fangxiao Decoction in treating asthma may be related with the up-regulation on IL-10 levels and downregulation on IL-17 A and CXCL10 levels. It could reduce the airway inflammation, so as to achieve the purpose of preventing and treating asthma.
