六君子汤对4-硝基喹啉-1-氧化物诱导食管癌小鼠免疫调节的影响 被引量:18

Effects of Liujunzi Decoction on quality of life and immune regulation of mice with esophageal carcinoma induced by 4NQO

摘要 目的:观察六君子汤对4-硝基喹啉-1-氧化物(4NQO)诱导食管癌小鼠一般情况、脏器重量和指数、食管组织病理形态、细胞因子及淋巴细胞亚群的干预作用。方法:小鼠分正常组、模型组和六君子组,16周后撤除干预因素,观察进食饮水并称重,32周后取脏器称重计算脏器指数,观察食管组织,检测外周血IL-10、TNF、IFN-γ、MCP-1、IL-6、IL-12p70等细胞因子、淋巴细胞亚群(CD3^+/CD19^+,CD3^+/CD8^+)。结果:六君子组小鼠活动度较正常,体质量下降不明显等;脾、心和肝重量高于模型组具有显著性差异(P<0.05),肾、肺指数显著降低(P<0.05);镜下食管基底细胞层未见"癌珠"和"病理分裂象",可上调IL-10、TNF、MCP-1、IL-6、IL-12p70(P<0.05),且显著提高CD4^+/CD8^+比例(P<0.05)。结论:六君子汤能改善4NQO诱导食管癌小鼠生存质量、保护脏器;防治食管组织损害;提高IL-10等细胞因子水平提升机体免疫功能,纠正CD4^+/CD8^+比例改善免疫功能失衡及免疫抑制。 Objective: To observe the effects of Liujunzi Decoction on the quality of life, weight, organ weight and organ index, esophageal pathomorphology, plasma cytokines and lymphocyte subgroup of mice with esophageal carcinoma induced by 4-Nitroquinoline 1-oxide(4NQO). Methods: All mice were divided into normal group, model group and Liujunzi Decoction group. After 16 weeks, all the intervention factors were removed, the diet and weight of mice were observed; After 32 weeks, all mice were sacrificed to get organ weight, to observe esophageal pathomorphology, to detect plasma cytokines such as IL-10, TNF, IFN-γ, MCP-1, IL-6, IL-12p70 and lymphocyte subgroup(CD3^+/CD19^+, CD3^+/CD8^+). Results: The activity of mice in Liujunzi Decoction group was normal, the declining of weight was not obvious; the weights of the spleen, heart, and liver were higher than the model group(P〈0.05), the organ index of kidney and lung were lower(P〈0.05), esophageal mucosa epithelial basal cell layer represented abnormal hyperplasia without "cancer pearl" and "pathological division image". Plasma cytokines IL-10, TNF, IFN-γ, MCP-1, IL-6, IL-12p70 were upregulated, CD4^+/CD8^+ ratio was increased. Conclusion: Liujunzi Decoction could improve the quality of life of the esophageal carcinoma mice, protect organs, prevent esophageal tissue damage, and improve the immune function by promoting IL-10 and other plasma cytokines and increasing CD4^+/CD8^+ ratio.
