高效液相色谱法同时测定杜仲叶中4种有效成分的含量 被引量:6

摘要 目的测定杜仲叶中4种有效成分的含量。方法采用HPLC测定绿原酸、槲皮素、山奈酚、芦丁的含量,色谱柱为Wonda Sil C_(18)Superb色谱柱(4.6 mm×250 mm,5μm),流动相:0.4%磷酸溶液(A)-甲醇(B)进行梯度洗脱,流速1 ml/min,进样量10μl,检测波长262 nm。结果绿原酸的质量在0.825~4.123μg(R2=0.9999)范围内呈良好线性关系,平均回收率为101.89%,RSD为0.49%;槲皮素的质量在0.165~0.824μg(R2=0.9998)范围内呈良好线性关系,平均回收率为99.88%,RSD为2.64%;山奈酚的质量在0.021~0.104μg(R^2=0.9999)范围内呈良好线性关系,平均回收率为100.0%,RSD为0.89%;芦丁的质量在0.002~0.012μg(R^2=0.9991)范围内呈良好线性关系,平均回收率为100.0%,RSD为0%。结论本方法简便、快速、准确、重复性好,适用于杜仲叶中各成分的含量测定。 Objective To determine the four effective components in Eucommia folium. Methods HPLC was employed to determine the contents of four effective components:chlorogenic acid,quercetin,kaempferol,rutin. The chromatogram colume was Wonda Sil C_(18)Superb(4.6 mm × 250 mm,5 μm),gradiently eluting with methanol as mobile phase A and 0.4% phosphoric acid as phase B at a flow rate of 1 ml/min. The injection volumn was 10 μl. Detection wavelength was set at 262 nm. Results Chlorogenic acid was linear in the range of 0.825-4.123 μg with correlation coefficient of 0.9999. The average recovery was 101.89% with RSD 0.49%(n=6). Quercetin was linear in the range of 0.165-0.824 μg with correlation coefficient of 0.9998. The average recovery was 99.88%with RSD 2.64%. Kaempferol was linear in the range of 0.021-0.104 μg with correlation coefficient of 0.9999. The average recovery was 100.0% with RSD 0.89%. Rutin was linear in the range of 0.002-0.012 μg with correlation coefficient of 0.9991. The average recovery was 100.0% with RSD 0%. Conclusion The method is convenient,fast,accurate and practicable. It is suitable for content determination of components in Eucommia folium.
