滩羊H-FABP基因序列测定及分析

Sequence determination and analysis of H-FABP gene in Tan Sheep

采集2、4、6、9、12、18月龄共计120只滩羊血液标本,PCR扩增H-FABP基因,进行酶切和测序.结果表明:H-FABP基因在滩羊群体中存在多态性,扩增的目的基因序列长度分别为285 bp、258 bp.通过测序发现外显子2在236、241、249处分别发生了A→C、C→T、T→G的单碱基突变.外显子3在103、164处分别发生了T→C、G→A的单碱基突变.

In order to study the genetic diversity of Heart type fatty acid binding protein（Heart, H- FABP） gene, A total of 120 blood specimens（ 2、4、6、9、12 and 18 months）of Tan sheep were collected.PCR amplification、enzyme digesting and sequencing of H-FABP gene were tested subsequently.The results show that there is H- FABP gene polymorphism in Tan sheep.The targen lengths were285 bp and 258 bp respectively.The sequencing test showed that A to C、C to T and T to G single base mutations respectively in the base of236, 241, 249 of exon 2,and T to C,G to A single-base mutation in the base of 103, 164 of exon 3.
