摘要
以四臂聚乙二醇为引发剂,辛酸亚锡为催化剂,通过开环聚合反应制备四臂星型聚乙二醇/聚乳酸共聚物。通过透析法制备了负载抗肿瘤药物羟基喜树碱的四臂星型聚乙二醇/聚乳酸纳米粒子,并对其粒径分布、载药量、稳定性、体外释药以及体外细胞毒性等特性进行了考察。结果表明,载药纳米粒子的粒径小于200 nm,粒径分布均匀,稳定性良好,在体外的药物缓释效果显著。体外细胞毒性实验结果表明,载药纳米粒子表现出优异的肿瘤细胞增殖抑制效果。
4-armed star poly( ethylene glycol) / polylactide copolymers( 4SPEG / PLA) were synthesized by ring-opening polymerization of lactide( LA) with 4-armed poly( ethylene glycol) as macromolecular initiator and stannous octoate as catalyst. Hydroxycamptothecin( HCPT)-loaded nanoparticles made from 4SPEG / PLA were prepared by dialysis method. The size and size distribution,drug loading content,stability,in vitro drug release and in vitro cell cytotoxicity of HCPT-loaded nanoparticles were studied.It was found that the size distribution was narrow and the average diameter was below 200 nm. In vitro drug release experiments indicated that HCPT-loaded nanoparticles showed sustained release profile. Cytotocicity tests showed that the cytotoxicity of HCPT-loaded nanoparticles against cancer cell in vitro was remarkably higher than that of free drugs. These data demonstrated that such HCPT-loaded nanoparticles may serve as a stable delivery system for poorly soluble HCPT.
