摘要
目的:建立来曲唑诱导的多囊卵巢综合征(polycystic ovarian syndrome,PCOS)大鼠模型,探讨复方麦芽丸(Compound Malt Pill)对PCOS大鼠卵巢组织形态及雄激素受体(Androgen Receptor,AR)、胰岛素受体(Insulin Receptor,INSR)表达的影响。方法:6周龄清洁级雌性SD大鼠按8ml/kg每天灌服0.125mg/ml来曲唑-羧甲基纤维素溶液,建立PCOS大鼠模型,成模大鼠随机分为5组:模型组、炔雌醇环丙孕酮1.44 g/kg组、复方麦芽丸5.76g/kg组、11.52 g/kg组、23.04 g/kg组,每组8只,分别给予相应浓度药物干预21d,另设同龄正常组大鼠8只。观察治疗后各组大鼠体重情况、卵巢形态大体变化,免疫组化、Western Blot检测AR、INSR蛋白表达水平。结果:(1)造模成功后,造模大鼠体重增加明显;药物干预治疗后,各治疗组与模型组比较体重变化不明显;(2)观察治疗后卵巢HE染色,与模型组比较卵泡囊状扩张数减少或缩小,颗粒细胞增多,原始卵泡增多,黄体增多;(3)ELISA结果:模型组睾酮(testosterone,T)、胰岛素(Insulin,INS)水平升高;复方麦芽丸各剂量组血清T、INS浓度较模型组明显降低,炔雌醇环丙孕酮1.44 g/kg组血清T值明显降低,而INS浓度较模型组差异不明显;(4)免疫组化和Western blot结果:模型组AR、INSR蛋白表达明显升高,复方麦芽丸5.76g/kg、11.52 g/kg、23.04 g/kg组均能下调AR、INSR表达。结论:复方麦芽丸可能通过调节AR、INSR水平,以改善PCOS模型大鼠异常的内分泌代谢状态、促进排卵。
Objective: To establish polycystic ovarian syndrome rat model induced by letrozole and investigate the effect of Compound Malt Pill on ovarian tissue morphology and the expression of AR,INSR in PCOS rats. Methods: The PCOS rat model was induced by 0. 125 mg / ml letrozole-CMC solution according to the dose of 8ml / kg at clean grade femal SD rats aged 6 weeks,after successful modeling randomly divided into5 groups( n = 8) : model group,Ethinylestradiol and Cyproterone Acetate Tablets 1. 44 g / kg dosed group,Compound Malt Pill 5. 76 g / kg dosed group,11. 52 g / kg dosed group,23. 04 g / kg dosed group,intervented by appropriate drug concentrations for 21 days respectively,another 8 rats were definited normal group. The rat body weight and change of ovarian morphology in every group were observed,and the expression of AR,INSR protein was examined by immunohistochemical and Western Blot. Results:( 1) After the successful modeling,the weight higher increased( compared with normal group,P 〈 0. 05); After intervented,the change of body weight did not change significantly( compared with model group,P 〉 0. 05).( 2) Observed ovary HE staining after treatment,compared with the model group,the number of follicular cystic dilatation was reduced,granulosa cells,primordial follicles and corpus luteum were increased.( 3) ELISA results: the level of T and INS in model group was increased( compared with normal group,P 〈 0. 05). The level of T and INS Compound Malt Pill 5. 76 g / kg,11. 52 g /kg and 23. 04 g / kg dosed groups were all decreased( P 〈 0. 05),the T level in Ethinylestradiol and Cyproterone Acetate Tablets 1. 44 g / kg dosed group was significantly drcreased,but the level of INS did not changed( P 〉 0. 05).( 4) Western blot and Immunohistochemistry results: the model group can significantly increase the expression of AR,INSR( P 〈 0. 05),and Compound Malt Pill 5. 76 g / kg,11. 52 g / kg and 23. 04 g / kg dosed groups can reduce the expression of AR and INSR( P 〈 0. 05). Conclusion: PCOS can be treated by Compound Malt Pill through regulating the level of AR,INSR,to improve the abnormal endocrine and metabolicstate and promote ovulation.
