补肾填精法改善糖尿病勃起功能障碍大鼠性功能的机制研究

Effect of jinshuibao capsule and metforformin on corpus cavernosum smooth muscle and eNOS and sexual function levels in Diabetic Erectile Dysfunction rats

目的观察补肾填精法对糖尿病勃起功能障碍(ED)大鼠性功能水平的影响及作用机制。方法采用SD大鼠腹腔注射链脲佐菌素法,造模后断尾取血检测血糖,当血糖大于16.6 mmol/L并颈项皮肤注射阿朴吗啡后无勃起,提示糖尿病勃起功能障碍大鼠模型造模成功。将造模成功的大鼠随机分为补肾填精法治疗组、阳性对照组和模型组各10只,同时留10只大鼠作为空白对照组,各组在给药8周时和停药2周后分别测定大鼠的性功能水平,同时检测大鼠阴茎组织形态学及一氧化氮合成酶(NOS)。结果补肾填精法治疗组大鼠阴茎组织平滑肌数量较模型组及阳性对照组有所增多,同时可使纤维密度降低及血管壁变薄。补肾填精法治疗组和阳性对照组NOS表达量分别为(64.1±5.91)和(34.0±4.81),与模型组(22.8±4.47)比较均有所提高,差异具统计学意义(P<0.01)。与空白对照组相比,模型组大鼠性功能显著下降,差异具统计学意义(P<0.01);补肾填精法治疗组和阳性对照组均可改善大鼠性功能,与模型组比较差异具统计学意义(P<0.01),补肾填精法治疗组远期疗效优于阳性对照组,差异具统计学意义(P<0.05)。结论补肾填精法能显著提高糖尿病勃起功能障碍大鼠的性功能并维持一定的时间,这可能与其能改善阴茎组织形态学及提高NOS表达水平有关。

Objective To investigate the influence of Jinshuibao Capsule and Metforformin on Corpus cavernosum smooth muscle and eNOS and Sexual function levels in Diabetic Erectile Dysfunction rats. Methods Rat model with DMED was established by injection with intraperitoneal STZ and Apomorhine. The established rat models were divided into four groups including Nourishing Kidney-essence group, melbine group, model group and normal group. The Phallic tissues and eNOS and sexual function levels of rats were measured in 8 weeks during drug administration and in 2 weeks after drug withdrawal. Results The amount of corpus cavernosum smooth muscle in Nourishing Kidney-essence group was higher than that in melbine group and model group, whereas fiber density and vessel wall decreased. The expression levels of eNOS in Nourishing Kidney-essence group and melbine group were all higher than that in modle group （both P〈0.01）, and the effect of Nourishing Kidney-essence group was more better. The level of sexual function in modle group was lower than that of the other. Nourishing Kidney-essence group and melbine group could improved sexual function and Nourishing Kidney-essence group was more better （P〈0.05）. Conclusion Nourishing Kidney-essence can signficantly upregulate the expressions of eNOS and improve sexual function level..
