摘要
目的:用酵母双杂交实验筛选人肝cDNA文库,得到与埃博拉病毒VP24、VP35、VP40蛋白相互作用的蛋白质,用以研究VP24、VP35、VP40蛋白的生物学功能。方法:以埃博拉病毒VP24、VP35、VP40为诱饵蛋白,利用酵母双杂交技术对人肝cDNA文库进行初步筛选,并采用DNA测序技术及生物信息学技术进行分析。结果:筛选到13个可能与VP24、VP35、VP40相互作用的蛋白,包括视黄酸受体应答蛋白2(RARRES2)、PRKA激酶相互作用蛋白1(AKIP1)、白蛋白(ALB)、补体因子1(CF1)、人肿瘤易感基因(TSG101)等。结论:应用酵母双杂交技术初步筛选出与埃博拉病毒VP24、VP35、VP40相互作用的宿主蛋白,并完成了初步功能分析。
Objective:In order to understand the biological function of VP24,VP35,VP40 proteins in Ebola virus,the bait plasmids of VP24,VP35,VP40 proteins were constructed and screened human liver eDNA library to obtain interactive proteins by yeast two-hybrid technique.Methods:Performed a preliminary screening for human liver eDNA library by yeast two-hybrid system,which based on Ebola virus VP24,VP35,VP40 proteins as baits,and analyzed results by the methods of DNA sequencing and bioinformatics techniques.Results:There were thirteen different proteins that may interact with VP24,VP35,VP40 had been screened out,including retinoic acid receptor responder protein 2(RARRES2),PRKA kinase interacting protein 1 (AKIP1),albumin(ALB),complement factor 1 (CF1),human tumor susceptibility gene (TSG101).Conclusion:Some proteins which may interact with Ebola virus VP24,VP35,VP40 have been screened out by yeast two-hybrid technique,and the initial function analysis has been completed.
