Effects of alpha-lipoic acid injection on oxidative stress in rats after cerebral ischemia reperfusion

摘要 目的探讨α-硫辛酸对大鼠脑缺血再灌注损伤大鼠氧化应激的影响。方法将90只SD大鼠随机分为假手术组、脑缺血再灌注组(模型组)和α-硫辛酸干预组,每组各30只。应用线栓法制备大脑中动脉闭塞2 h后再灌注模型,分别于再灌注6 h、24 h、3 d、7 d采用2,3,5-氯化三苯基四氮唑(TTC)染色法检测脑梗死灶大小,对其神经功能障碍进行评分,检测脑组织中丙二醛(MDA)、超氧化物歧化酶(SOD)、一氧化氮(NO)含量及诱导型一氧化氮合酶(i NOS)活性。结果与模型组比较,α-硫辛酸干预组大鼠脑梗面积缩小,脑组织中的MDA、NO含量及i NOS活性均显著降低,SOD活性显著升高(P<0.05)。结论α-硫辛酸对大鼠脑缺血再灌注损伤具有一定的保护作用,其机制可能是α-硫辛酸清除自由基抗氧化作用。 Objective To explore the effects of alpha-lipoic acid injection on oxidative stress in rats after cerebral ischemia reperfusion. Methods The 90 male Sprague-Dawley( SD) rats were randomly divided into the sham-operation group,the cerebral ischemiareperfution group( model group) and the alpha-lipoic acid pretreatment ischemia-reperfution group,with 30 in each group. A model of middle cerebral artery( MCA) occlusion for 2 hours followed by reperfution was induced in rats using sutures methold. The infarct volume of the brain was determined by 2,3,5-triphenyltetrazolium chloride( TTC) staining at 24 hours after reperfution,respectively.Scores of neurological dysfunction,the malondialdehvde( MDA),superoxide dismutase( SOD),nitric oxide( NO) levels in brain tissues,and the activity of inducible NO synthase( i NOS) were detected. Results The scores of neurological dysfunction,cerebral infarction size,contents of MDA and NO,and the activity of i NOS in alpha-lipoic acid group were significantly reduced than those in the model group,while the activity of SOD in alpha-lipoic acid group was significantly increased than that in the model group( P〈0. 05).Conclusion Alpha-lipoic acid injection has the protective effect on focal cerebral ischemia reperfusion injury,and the mechanism may be related to scavenging free radicals.
