摘要
研究建立了鸡血浆中替米考星的高效液相色谱(HPLC)检测方法。血浆中替米考星采用甲醇和三氯甲烷提取,流动相为乙腈:四氢呋喃:二丁胺磷酸缓冲液:超纯水(110:55:25:810,v/v),紫外检测波长286nm,流速1mL/min。结果显示,替米考星标准工作液在O.05。10mg/L范围内呈良好线性关系(R^2≥0.9998)。血浆中替米考星添加浓度为5.0、0.5、0.05mg/L时,回收率为97.0%~99.8%;日内和日间变异系数分别小于7.2%和7.7%。该方法前处理简单省时,不用固相萃取小柱净化,节约试验成本,为替米考星的检测提供了方法学基础。
This study established a high performance liquid chromatography (HPLC) method for determination of tilmicosin in plasma. Methanol and chloroform were used for extraction of tilmicosin from plasma. Mobile phase was as following: acetonitrile,tetrahydrofuran,butyl amine phosphate buffer,uhrapure water with the proportion of 110:55:25:810 (v/v). UV detection with wavelength of 286 nm was used for detector, the flow rate was 1 mL/min. Tilmicosin standard working solution within the scope of 0.05 to 10 mg/L showed good linear relationship (R2≥0.9998). The recovery rate was 97.0% to 99.8% at concentration of 5.0,0.5,0.05 mg/L. The intra-day and inter-day coefficient of variation were less than 7.2% and 7.7% respectively. Pre-treatment of the method was simple, timesaving and cost-effective without use of solid phase extraction column,which provided methodology basis for tilmicosin determination.
