摘要
为研究复合酶辅助高压法提取金针菇黄酮的最适条件,本试验通过单因素试验和正交试验,对固液比、酶反应时间、酶反应温度、酶反应pH、高压时间及高压温度对黄酮得率的影响进行了研究,并以金针菇黄酮得率为评价指标,优化提取工艺。试验结果表明,复合酶辅助高压法提取金针菇黄酮的最优条件是:固液比为1∶35(g/m L),酶反应时间为80 min,酶反应温度为50℃,酶反应pH为6,高压处理温度为115℃,高压处理时间为40 min。以上述条件从金针菇中提取黄酮,得率可达13.45%。
In this study,a composite enzyme assisted with high pressure extraction method was developed for the ex-traction of flavonoids from Flammulina velutiper.The extraction conditions were optimized through the single factor and or-thogonal tests.The effects of solid-liquid ratio,enzymatic reaction time,enzymatic reaction temperature,enzymatic reaction pH,high-pressure extraction time and high-pressure extraction temperature on the extraction yield of flavonoids were in-vestigated.The experimental results showed that the optimal condition was solid-liquid ratio of 1∶35(g/mL),enzymatic reac-tion time of 80 min,enzymatic reaction temperature of 50℃,enzymatic reaction pH of 6,high-pressure extraction temper-ature of 115 ℃ and hig-pressure extraction time of 40 min.Under these conditions,the extraction yield of flavonoids from Flammulina velutipe was up to 13.45%.
